Discovery of Ro 41‐0960, a Non‐Steroidal Inhibitor of the Na+/K+‐ATPase [PDF]

open access: yesChemMedChem, Volume 21, Issue 17, 14 September 2026.
Ro 41‐0960 is a non‐steroidal Na+/K+‐ATPase inhibitor with low micromolar potency and a mixed inhibitory mechanism distinct from classical cardiotonic steroids, without acute pro‐arrhythmic effects in human cardiomyocytes. The Na+/K+‐ATPase (NKA) is a central regulator of cardiac ion homeostasis and a validated target for heart failure.

Modular Total Synthesis and Cell-Based Anticancer Activity Evaluation of Ouabagenin and Other Cardiotonic Steroids with Varying Degrees of Oxygenation. [PDF]

open access: yesJ Am Chem Soc, 2019
A Cu(II)-catalyzed diastereoselective Michael/aldol cascade approach is used to accomplish concise total syntheses of cardiotonic steroids with varying degrees of oxygenation including cardenolides ouabagenin, sarmentologenin, 19-hydroxysarmentogenin ...

Na/K-ATPase in the renal proximal tubule cell: classic & novel roles in sodium handling [PDF]

open access: yesFrontiers in Physiology
The cell membrane Na/K-ATPase (NKA), the archetypal P-type ATPase, is the enzymatic complex that hydrolyzes ATP to transport Na+ and K+ against their transmembrane electrochemical gradients.

Neuroinflammation and Neutrophils: Modulation by Ouabain

open access: yesFrontiers in Pharmacology, 2022
Cardiotonic steroids are natural compounds that present many physiological and pharmacological functions. They bind Na+/K+-ATPase (NKA) modifying cellular ion concentration and trigger cell signaling mechanisms without altering ion balance.

Cannogenol and Related Cardiotonic Steroids: Concise Synthesis and their Anticancer Activity Evaluation [PDF]

open access: yes, 2018
Small molecules that are approved as drugs are largely the result of extensive synthetic efforts. A robust synthetic method is required to access the natural products and their analogs to study their biological activities.

Unveiling the Role of Cholesterol in Subnanomolar Ouabain Rescue of Cortical Neurons from Calcium Overload Caused by Excitotoxic Insults

open access: yesCells, 2023
Na/K-ATPase maintains transmembrane ionic gradients and acts as a signal transducer when bound to endogenous cardiotonic steroids. At subnanomolar concentrations, ouabain induces neuroprotection against calcium overload and apoptosis of neurons during ...

Effect of ouabain on calcium signaling in rodent brain: A systematic review of in vitro studies

open access: yesFrontiers in Pharmacology, 2022
The Na+/K+-ATPase is an integral membrane ion pump, essential to maintaining osmotic balance in cells in the presence of cardiotonic steroids; more specifically, ouabain can be an endogenous modulator of the Na+/K+-ATPase. Here, we conducted a systematic

Steroids—has the time come to extend their use to AML?

open access: yesJournal of the Egyptian National Cancer Institute, 2021
Background In 2018, leukaemia accounted for 2.6% of all new cancers, it being the 13th most common cause of cancer and the 10th most common cause of cancer death. Glucocorticoids are commonly used in lymphoid leukaemia treatment, where they are cytotoxic.

Na/K-ATPase amplification of oxidant stress; a universal but unrecognized clinical target?

open access: yesMarshall Journal of Medicine, 2016
The Na/K-ATPase has a signaling function which appears to be separate from its ion pumping function. This signaling function refers to the transduction of conformational changes in the Na/K-ATPase alpha1 subunit into activating Src’s tyrosine kinase ...
