Synergistic Integration of Recombinase Polymerase Amplification and CRISPR–Cas12a Enables High‐Sensitivity Detection of Mycoplasma bovis

open access: yesMicrobiologyOpen, Volume 15, Issue 5, October 2026.
This graphical abstract illustrates a rapid diagnostic platform for Mycoplasma bovis detection using Recombinase Polymerase Amplification coupled with CRISPR–Cas12a targeting the uvrC gene. The assay operates at 37°C–39°C, delivers results within 1 h, and demonstrates complete concordance with PCR in cattle and bison clinical samples.

Targeting of CRISPR-Cas12a crRNAs into human mitochondria

open access: yesBiochimie
Mitochondrial gene editing holds great promise as a therapeutic approach for mitochondrial diseases caused by mutations in the mitochondrial DNA (mtDNA). Current strategies focus on reducing mutant mtDNA heteroplasmy levels through targeted cleavage or base editing. However, the delivery of editing components into mitochondria remains a challenge. Here

Graft Biology in the CRISPR Era: From Tissue Fusion to Genome Compatibility

open access: yesPlant-Environment Interactions, Volume 7, Issue 5, October 2026.
ABSTRACT Plant lineage has traditionally constrained grafting compatibility, with monocots generally considered incompatible because of their dispersed vascular bundles and limited secondary growth. Recent studies have shown that embryonic grafting can establish successful graft unions in selected monocot systems by exploiting early developmental ...

Development of a Recombinase Polymerase Amplification and CRISPR-Cas12a-Based Assay for Rapid Detection of Rice Bakanae Disease Caused by Fusarium fujikuroi

open access: yesAgronomy
Fusarium fujikuroi is the primary causal agent of rice bakanae disease, which can lead to substantial yield losses. Developing a rapid, highly specific, and accurate method for detecting F. fujikuroi is crucial for effective surveillance, prevention, and

A dual gating mechanism controls target‐strand cleavage in Cas12j: Implications for engineering efficient nickases

open access: yesProtein Science, Volume 35, Issue 10, October 2026.
Abstract The rapid expansion of CRISPR technologies has unveiled a diverse repertoire of RNA‐guided endonucleases, among them the compact Cas12j, which has emerged as a promising genome‐editing tool. Cas12j cleaves the two DNA strands sequentially; however, the molecular mechanism that regulates this cleavage remains incompletely understood.

A Robust Framework for Maize Elite Line Genome Editing Through Enhanced HI‐Edit via LbCas12a Activity Optimization

open access: yesPlant Biotechnology Journal, Volume 24, Issue 10, Page 5392-5404, October 2026.
ABSTRACT Haploid induction coupled with genome editing (HI‐Edit) enables direct modification of commercial crop varieties, bypassing the need for trait introgression or direct transformation of elite lines with CRISPR machinery. However, its widespread application has been constrained by low haploid editing rates (HER), the proportion of haploids ...

CRISPR-Cas12a–assisted PCR tagging of mammalian genes

open access: yesJournal of Cell Biology, 2018
Here we describe a time-efficient strategy for endogenous C-terminal gene tagging in mammalian tissue culture cells. An online platform is used to design two long gene-specific oligonucleotides for PCR with generic template cassettes to create linear dsDNA donors, termed PCR cassettes.

Generation of Allogeneic CAR‐T Circumvents Functional Deficits in Patient‐Derived Autologous Product for Glioblastoma

open access: yesInternational Journal of Cancer, Volume 159, Issue 6, Page 1521-1534, 15 September 2026.
Clinical trials of chimeric antigen receptor T‐cell (CAR‐T) therapies in glioblastoma have shown limited clinical benefits. Whether this may be explained by the basal quality of CAR‐T products, which are currently generated using patient, autologous T‐cells, has been little explored.
