Genetic risk factors associated with ocular perfusion pressure in primary open-angle glaucoma

open access: yesHuman Genomics
Background Primary open-angle glaucoma (POAG) is the leading cause of irreversible vision loss. However, its genetic risk factors, such as the vascular hypothesis of POAG, remain unclear.

Causal mediation analysis in instrumental-variables regressions [PDF]

open access: yes
In this article, we describe the use of ivmediate, a new command to estimate causal mediation effects in instrumental-variables settings using the framework developed by Dippel et al. (2020, unpublished manuscript).

Dimethyl fumarate combined with cisplatin at subcytotoxic doses sensitizes cervical cancer toward ferroptosis and apoptosis through GSH restriction and p53 (re)activation

open access: yesMolecular Oncology, EarlyView.
Dimethyl fumarate (DMF) reduces growth of HPV‐positive cervical cancer spheroids and induces ferroptosis in cervical cancer cells via blocking SLC7A11/Glutathione (GSH) axis. Combination of subcytotoxic doses of DMF and cisplatin (CDDP) further suppresses spheroid growth and drives cell death in 2D culture models.

A comparison and evaluation of statistical methods for mediation analysis with mixtures of environmental exposures

open access: yesBMC Medical Research Methodology
Background Environmental studies often evaluate how exposures influence health outcomes through intermediate biological processes. In practice, researchers are often interested in complex exposure mixtures rather than single agents, creating challenges ...

Methods in causal inference. Part 2: Interaction, mediation, and time-varying treatments

open access: yesEvolutionary Human Sciences
The analysis of ‘moderation’, ‘interaction’, ‘mediation’ and ‘longitudinal growth’ is widespread in the human sciences, yet subject to confusion. To clarify these concepts, it is essential to state causal estimands, which requires the specification of ...

Mediation Analysis in Experimental Research

open access: yes, 2018
This chapter introduces the conceptual and statistical basics of mediation analysisin the context of experimental research. Adopting the respective terminology, mediation analysis can be referred to as an array of quantitative methods developedto ...

Network divergence analysis identifies adaptive gene modules and two orthogonal vulnerability axes in pancreatic cancer

open access: yesMolecular Oncology, EarlyView.
Tumors contain diverse cellular states whose behavior is shaped by context‐dependent gene coordination. By comparing gene–gene relationships across biological contexts, we identify adaptive transcriptional modules that reorganize into distinct vulnerability axes.

Causal Mediation Analysis with Spatial Interference

open access: yes
In the causal inference framework, estimating causal effects requires specific assumptions, among which is the assumption of independence of each unit’s outcome from the treatment assigned to other units.

COMP–PMEPA1 axis promotes epithelial‐to‐mesenchymal transition in breast cancer cells

open access: yesMolecular Oncology, EarlyView.
This study reveals that cartilage oligomeric matrix protein (COMP) promotes epithelial‐to‐mesenchymal transition (EMT) in breast cancer. We identify PMEPA1 (protein TMEPAI) as a novel COMP‐binding partner that mediates EMT via binding to the TSP domains of COMP, establishing the COMP–PMEPA1 axis as a key EMT driver in breast cancer.

Causal relationships between gut microbiota, plasma metabolites, and HIV infection: insights from Mendelian randomization and mediation analysis

open access: yesVirology Journal
Objective Gut dysbiosis and metabolic abnormalities have been implicated in HIV infection. However, the exact causal relationships among the gut microbiota, metabolites, and HIV infection remain poorly understood.
