Genetic analysis of the Drosophila cdc2 homolog

ABSTRACT We have identified mutations in the Drosophila cdc2 gene. The recessive lethality of these mutant alleles was rescued after P-element-mediated transformation with a genomic cdc2 fragment. Sequence analysis of amorphic alleles revealed non-conservative exchanges in evolutionary conserved positions.

CDK9 (PITALRE): A multifunctional cdc2-related kinase

CDK9 is a cdc2-related kinase protein. Previously named PITALRE, this protein is a serine-threonine kinase involved in many physiological processes. Unlike most of the cdc2-like kinases, its activity is not cell cycle-regulated. CDK9 acts preferentially in processes different from cell-cycle regulation, such as differentiation.

Synthetic peptide representing the site Ser-41 in vimentin, Leu-Gly-Ser41-Ala42-Leu-Arg44-Arg-Arg-NH2, and its analogs in which Ala-42 was replaced by various amino acids were tested as substrates for cdc2 kinase. Among them, the analog containing sarcosine as well as proline was an excellent substrate.

Beacon interacts with cdc2/cdc28-like kinases

Previously we found elevated beacon gene expression in the hypothalamus of obese Psammomys obesus. Beacon administration into the lateral ventricle of P. obesus stimulated food intake and body weight gain. In the current study we used yeast two-hybrid technology to screen for proteins in the human brain that interact with beacon.

Demonstration of cdc2 kinase in mature neuronal nuclei

Immunocytochemistry has been used to assess the distribution of the mitosis-associated protein kinase, cdc2, in mature neurons of rat brain. A robust signal is apparent in most, but not all neurons in cerebellum, cortex and hippocampus. The signal is concentrated in neuronal nuclei with lower levels of immunoreactivity in neuronal cytoplasm.

Nuclear factor I (NF-I) was purified to near homogeneity from Ehrlich ascites tumor cells. Mouse NF-I consisted of two peptides with relative molecular masses of 53 and 55 kDa and bound to the authentic NF-I site of the adenovirus DNA with no significant affinity with the CCAAT box.
