100歳から115歳にわたる5例の長寿者の脳を病理学的に検討した.1) 症例はいずれも女性で, 従来ほとんど病気らしい病気をせず, 比較的長寿の家系のものが多い.2) 精神機能の障害は死亡の数年以内に目立ってきている.3) 死因は, 肺炎, 心不全, 肺癌, その他である.4) 全身臓器の著明な萎縮があり, 脳重は950~1,170gで, 全葉に回転萎縮がみられた.5) 脳に肉眼的粗大病変のあるものは3例で, 出血, 梗塞とも小さなものであった.6) 組織学的には, 生理的老年変化と関連した神経細胞数の減少, 萎縮, リポフスチン沈着, 軸索ジストロフィー, マリネスコ小体, トルペドーなどはいずれも高度であった.

Such nutritional phenotyping studies can bridge the gap between in vitro studies and in vivo research in mammals.

This schematic illustrates the hierarchical and interconnected nature of the primary molecular hallmarks of aging. The progression of aging is driven by a convergence of intrinsic molecular insults. Within the nucleus, genomic instability and telomere attrition trigger persistent DDR, accompanied by extensive epigenetic alterations.
