Human anti‐α‐fucose antibodies are xenoreactive toward GGTA1/CMAH knockout pigs
Xenotransplantation, 2020AbstractProgress has been made in overcoming antibody‐mediated rejection of porcine xenografts by deleting pig genes that produce unique carbohydrate epitopes. Pigs deficient in galactose α‐1,3 galactose (gene modified: GGTA1) and neu5Gc (gene modified: CMAH) have reduced levels of human antibody binding.

AimCytidine monophosphate‐N‐acetylneuraminic acid (Neu5Ac) hydroxylase (Cmah) is an enzyme, which converts Neu5Ac to the sialic acid Neu5Gc. Neu5Gc is thought to increase inflammatory cytokines, which are, in part, produced in senescent cells of adipose tissues.

Chinese hamster ovary (CHO) cells are the most widely used platform for recombinant therapeutic protein (RTP) production. Traditionally, the development of CHO cell lines has mainly depended on random integration of transgenes into the genome, which is not conducive to stable long-term expression.

Bioprosthetic heart valves (BHVs) originating from pigs are extensively used for heart valve replacement in clinics. However, recipient immune responses associated with chronic calcification lead to structural valve deterioration (SVD) of BHVs. Two well-characterized epitopes on porcine BHVs have been implicated in SVD, including galactose-α1,3 ...

Abstract Abstract 3773 Adeno-Associated Virus is a popular gene transfer vector that successfully treats many genetic diseases in animal models, yet has limited proof of efficacy in humans. While experimental animals have been useful in predicting vector dosing, they failed to provide a model for human T cell responses to ...
