Effects of synthetic cohesin-containing scaffold protein architecture on binding dockerin-enzyme fusions on the surface of Lactococcus lactis

open access: yesMicrobial Cell Factories, 2012
Background The microbial synthesis of fuels, commodity chemicals, and bioactive compounds necessitates the assemblage of multiple enzyme activities to carry out sequential chemical reactions, often via substrate channeling by means of multi-domain or ...

Cohesin residency determines chromatin loop patterns

open access: yeseLife, 2020
The organization of chromatin into higher order structures is essential for chromosome segregation, the repair of DNA-damage, and the regulation of gene expression.

The organizer of chromatin topology RIF1 ensures cellular resilience to DNA replication stress

open access: yesLife Science Alliance, 2023
The conserved protein RIF1 stabilizes replication factories and regulates cohesin recruitment in response to replication stress to limit DNA lesions. Eukaryotic genomes are duplicated from thousands of replication origins that fire sequentially forming a

ATZ‐1 promotes DNA replication efficiency to maintain normal meiotic function

open access: yesFEBS Open Bio, EarlyView.
Absence of ATZ‐1 interferes with meiotic DNA replication and cell cycle function via CHK‐1. This causes downstream defects associated with DNA damage and genomic integrity. Taken together, this study suggests that ATZ‐1 influences DNA replication efficiency and cell cycle function to maintain normal meiotic function.

Cohesin codes – interpreting chromatin architecture and the many facets of cohesin function [PDF]

open access: yesJournal of Cell Science, 2013
Summary Sister chromatid tethering is maintained by cohesin complexes that minimally contain Smc1, Smc3, Mcd1 and Scc3. During S-phase, chromatin-associated cohesins are modified by the Eco1/Ctf7 family of acetyltransferases. Eco1 proteins function during S phase in the context of replicated sister chromatids to convert chromatin-bound ...

Elaborate cellulosome architecture of Acetivibrio cellulolyticus revealed by selective screening of cohesin–dockerin interactions [PDF]

open access: yesPeerJ, 2014
Cellulosic waste represents a significant and underutilized carbon source for the biofuel industry. Owing to the recalcitrance of crystalline cellulose to enzymatic degradation, it is necessary to design economical methods of liberating the fermentable ...

Dual native G‐quadruplex folding is associated with chromatin looping at the MYC locus

open access: yesFEBS Open Bio, EarlyView.
BG4‐detectable G‐quadruplex (G4) in HaCaT and NHEK keratinocytes identified folded and unfolded G4s enriched at promoters/TSSs and active enhancers, whereas unfolded G4s also overlapped weak/poised enhancers. At MYC–PVT1, 3C‐qPCR detected enhancer–promoter looping only when G4s were simultaneously folded at both regulatory elements under native ...

Intermediate step of cohesin’s ATPase cycle allows cohesin to entrap DNA [PDF]

open access: yesProceedings of the National Academy of Sciences, 2018
Significance Cohesin is an ATPase that organizes chromosome structure for chromosome segregation, gene expression, and DNA repair. Its function has been implicated in the prevention of cancer, birth defects, and several human disorders.
