Complement Factor H: solution structures and interactions with ligands

open access: yes, 2009
Factor H (FH) is a plasma glycoprotein that plays a central role in regulating the alternative pathway of complement. It is composed of 20 short complement regulator (SCR) domains.

Tumor‐Intrinsic ARHGEF3 Enhances Antitumor Immunity by Promoting T‐Cell Infiltration and Limiting Myeloid Cell‐Mediated Immunosuppression

open access: yesAdvanced Science, EarlyView.
ARHGEF3 is broadly downregulated across human cancers and correlates with patient prognosis. Tumor‐intrinsic ARHGEF3 activates the RHOA–ROCK–PTEN cascade to inhibit AKT signaling, thereby promoting chemokine‐driven T‐cell infiltration and relieving lipid‐mediated myeloid immunosuppression.

A case of remittent C1-inhibitor deficiency

open access: yes, 2015
C1-inhibitor is a serine protease inhibitor (serpin) controlling complement and kinin/contactsystem activation. Mutations in C1-inhibitor gene almost consistently result in reduced C1-inhibitor functional level in plasma causing hereditary angioedema, a ...

Introgressed Variation in TaMYB7‐A1 Drives Graded Dormancy and Climate‐Adaptive Pre‐Harvest Sprouting Resistance in Wheat

open access: yesAdvanced Science, EarlyView.
TaMYB7‐A1 directly activates TaABI5 to enhance ABA signaling and regulate ABA‐GA homeostasis, enforcing seed dormancy. Its superior allele, derived from wild einkorn introgression, harbors a MITE insertion that elevates expression and two amino acid substitutions that enhance transcriptional activity, collectively generating graded PHS resistance for ...

Upregulation of C1-inhibitor in pancreatic cancer

open access: yes, 2019
Purpose: The complement system has recently sparked more interest in cancer research. The classical pathway is initiated by activation of the C1 complex, which irreversibly can be bound to and inhibited by C1-INH.

A Computational Strategy for Identifying Self‐Assembling Food‐Derived Molecules for Antiparasitic Nanotherapy

open access: yesAdvanced Science, EarlyView.
An integrated computational screening strategy identified ursolic acid (UA) and 18β‐glycyrrhetinic acid (18βGA) as a self‐assembling food‐derived molecular pair. The resulting carrier‐free nanoparticles (UA‐18βGA) showed synergistic antiparasitic activity, reduced combined toxicity, and host‐protective anti‐inflammatory effects in zebrafish and murine ...

Complement Activation During Ischemia/Reperfusion Injury Induces Pericyte-to-Myofibroblast Transdifferentiation Regulating Peritubular Capillary Lumen Reduction Through pERK Signaling

open access: yesFrontiers in Immunology, 2018
Pericytes are one of the principal sources of scar-forming myofibroblasts in chronic kidneys disease. However, the modulation of pericyte-to-myofibroblast transdifferentiation (PMT) in the early phases of acute kidney injury is poorly understood.

The Landscape and Regulation of Histone Crotonylation in Mammalian Gametes and Early Embryos

open access: yesAdvanced Science, EarlyView.
Histone crotonylation undergoes a genome‐wide transition from broad domains to canonical narrow peaks during minor zygotic genome activation (ZGA). This remodeling is required for proper major ZGA and blastocyst formation. Disruption of this transition by transcriptional inhibition, metabolic perturbation, or HDAC1 dysfunction impairs embryonic ...

Molecular mechanism for the pathogenicity of the human C1-inhibitor Arg378Cys variant

open access: yes, 2007
C1-inhibitor (C1-inh) is a member of serine protease inhibitor (serpin) family which contains a common tertiary structure composed of three -sheets and several α-helices. Genetic variants of C1-inh lead to plasma deficiency, causing Hereditary Angioedema

Cancer‐Associated Fibroblast‐Derived Sphingosine‐1‐Phosphate Activates a MALL–SDC4 Axis to Facilitate Perineural Invasion in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
CAF‐derived S1P activates the S1PR3‐JNK‐Jun signaling pathway, thereby transcriptionally upregulating MALL expression in cancer cells. MALL increases the cell‐surface abundance of SDC4 by promoting its recycling and restricting its lysosomal degradation, which in turn enhances RhoA‐p‐MLC2‐dependent amoeboid invasion and SDC4‐PTN axis‐mediated cancer ...
