In vitro models of cancer‐associated fibroblast heterogeneity uncover subtype‐specific effects of CRISPR perturbations

open access: yesMolecular Oncology, EarlyView.
Development of therapies targeting cancer‐associated fibroblasts (CAFs) necessitates preclinical model systems that faithfully represent CAF–tumor biology. We established an in vitro coculture system of patient‐derived pancreatic CAFs and tumor cell lines and demonstrated its recapitulation of primary CAF–tumor biology with single‐cell transcriptomics ...

Transcriptional network analysis of PTEN‐protein‐deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication

open access: yesMolecular Oncology, EarlyView.
Combining PTEN protein assessment and transcriptomic profiling of prostate tumors, we uncovered a network enriched in senescence and extracellular matrix (ECM) programs associated with PTEN loss and conserved in a mouse model. We show that PTEN‐deficient cells trigger paracrine remodeling of the surrounding stroma and this information could help ...

Spiking neural networks for continuous control via end-to-end model-based learning

open access: yesNeuromorphic Computing and Engineering
Despite recent progress in training spiking neural networks (SNNs) for classification, their application to continuous motor control remains limited. Here, we demonstrate that fully spiking architectures can be trained end-to-end to control robotic arms ...

Thermodynamics of continuous non-Markovian feedback control

open access: yesNature Communications, 2020
The second law of thermodynamics cannot be straightforwardly applied to systems with non-Markovian feedback control, and previous extensions to cover that regime have not been tested experimentally.

Deliberate Control of Continuous Motor Performance

open access: yesJournal of Motor Behavior, 2005
The authors examined the means by which people vary movement parameters to satisfy more than 1 constraint at a time in a repetitive motor task. The authors expected that when participants (N = 12) were simultaneously confronted with spatial and temporal constraints in an ellipse-drawing task, they would either exploit the intrinsic amplitude-frequency ...

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Continuous Sampling Procedures Without Control

open access: yesThe Annals of Mathematical Statistics, 1959
1. Summary. Several modifications of the Dodge CSP-1 procedure [1] are presented. Changes are made in the rule of action when a defective item is observed while on sampling. The Average Outgoing Quality Limit (AOQL) for these new procedures are derived without the assumption of control. These results are compared with the AOQL assuming control.

Recurrent cancer‐associated ERBB4 mutations are transforming and confer resistance to targeted therapies

open access: yesMolecular Oncology, EarlyView.
We show that the majority of the 18 analyzed recurrent cancer‐associated ERBB4 mutations are transforming. The most potent mutations are activating, co‐operate with other ERBB receptors, and are sensitive to pan‐ERBB inhibitors. Activating ERBB4 mutations also promote therapy resistance in EGFR‐mutant lung cancer.
