RdRp (RNA-dependent RNA polymerase): A key target providing anti-virals for the management of various viral diseases [PDF]

open access: yes, 2022
With the arrival of the Covid-19 pandemic, anti-viral agents have regained center stage in the arena of medicine. Out of the various drug targets involved in managing RNA-viral infections, the one that dominates almost all RNA viruses is RdRp (RNA ...

Genomic Characterization of Severe Acute Respiratory Syndrome-Related Coronavirus in European Bats and Classification of Coronaviruses Based on Partial RNA-Dependent RNA Polymerase Gene Sequences [PDF]

open access: yesJournal of Virology, 2010
ABSTRACT Bats may host emerging viruses, including coronaviruses (CoV). We conducted an evaluation of CoV in rhinolophid and vespertilionid bat species common in Europe. Rhinolophids carried severe acute respiratory syndrome (SARS)-related CoV at high frequencies and concentrations (26% of animals are positive; up to 2.4 × 10

Emerging RNA-Dependent RNA Polymerase Mutation in a Remdesivir-Treated B-cell Immunodeficient Patient With Protracted Coronavirus Disease 2019

open access: yesClinical Infectious Diseases, 2020
Abstract Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a newly discovered virus for which remdesivir is the only antiviral available. We report the occurrence of a mutation in RdRP (D484Y) following treatment with remdesivir in a 76-year-old female with post-rituximab B-cell immunodeficiency and persistent SARS-CoV-2 ...

Identifying transcription factors controlling the basal expression of human MRP4 highlights a substantial role for Sp1

open access: yesFEBS Open Bio, EarlyView.
The MRP4 transporter exports several drugs and signaling molecules. Here, we identified key promoter elements regulating basal MRP4 expression. Using reporter assays, we defined a conserved region with essential Sp1 and contributory Ets sites, which controlled basal MRP4 expression.

Replication enhancer elements within the open reading frame of tick-borne encephalitis virus and their evolution within the Flavivirus genus [PDF]

open access: yes, 2011
We provide experimental evidence of a replication enhancer element (REE) within the capsid gene of tick-borne encephalitis virus (TBEV, genus Flavivirus).

Sensitive and Spatially‐Resolved Electrochemiluminescence via Micropatterning

open access: yesAdvanced Materials Interfaces, EarlyView.
The use of a simple silica nanoparticle surface coating and patterning process can transform ordinary gold electrodes into multiplexed ECL sensing electrodes. Through spatially confined droplet formation and local ECL emission, it is possible to perform multiple analyses simultaneously from individual locations as well as intensify the ECL signal by as

Secreted Nonstructural Protein 3 is a Pathogenic Determinant of Orbivirus

open access: yesAdvanced Science, EarlyView.
This study uncovers a conserved PIP2‐dependent secretory pathway of orbivirus NS3 that induces vascular leakage. Pharmacological disruption of PIP2‐NS3 interaction significantly reduces viral pathogenicity and provides protective efficacy in murine models, establishing PIP2‐mediated NS3 secretion as both a key virulence determinant and a promising ...

ZDHHC18‐Mediated Palmitoylation of ORF3a Promotes SARS‐CoV‐2 Pathogenesis by Antagonizing TRIM16‐Mediated Ubiquitination and Proteasomal Degradation

open access: yesAdvanced Science, EarlyView.
Palmitoylation by ZDHHC18 blocks ORF3a K27‐linked ubiquitination mediated by TRIM16, thereby preventing its proteasomal degradation and strengthening viral pathogenesis. Targeting palmitoylation through a pharmacological inhibitor (2‐BP), a competitive inhibitory peptide (OPIP), or adenovirus‐mediated knockdown of ZDHHC18 expression presents a ...
