Cellular Requirements for CRM1 Import and Export

Journal of Biochemistry, 2003
We studied the cellular requirements for the translocation of CRM1 (exportin 1) between the nucleus and the cytoplasm. CRM1 import requires neither ATP, Ran, Ran-dependent GTP hydrolysis, nor a particular temperature. CRM1 and importin beta compete with each other during their import.

CRM1 expression in pancreatic carcinoma.

Journal of Clinical Oncology, 2013
e15115 Background: Increased expression of chromosomal region maintenance (CRM1) protein, also known as exportin 1, has been described in several human cancers. It is an important regulator of subcellular localization of tumor suppressor proteins and growth regulatory proteins. Direct inhibition of CRM1 blocks cell proliferation and induces apoptosis

ZAP is a CRM1-dependent nucleocytoplasmic shuttling protein

Biochemical and Biophysical Research Communications, 2004
The zinc finger antiviral protein (ZAP) is a recently isolated host antiviral factor. It specifically inhibits the replication of Moloney murine leukemia virus (MMLV) and Sindbis virus (SIN) by preventing the accumulation of viral RNA in the cytoplasm.

CRM1 mediates the nuclear export of YTHDF2

Biochemical and Biophysical Research Communications
Precise intracellular localization is crucial for protein function. YTHDF2, a reader protein for N6-methyladenosine (m6A), has been reported to shuttle between the cytoplasm and nucleus through an unknown mechanism. Here, we identify a functional nuclear export sequence (NES) in YTHDF2 that mediates its nuclear export.

Regulation of Crm1 expression in cancer cells

Clinical Cancer Research, 2010
Abstract Crm1 (or exportin 1) is a member of the karyopherin β protein family that mediates the nuclear export of cargo proteins and certain RNAs from the nucleus into the cytoplasm. We previously identified increased expression of Crm1 mRNA and protein in cervical tumours compared to normal epithelium, and increased Crm1 expression in ...

CRM1 as a new therapeutic target for non-Hodgkin lymphoma

Leukemia Research, 2015
The chromosomal region maintenance 1 (CRM1) may serve as a novel target for cancer treatment. Here, we investigated the anti non-Hodgkin lymphoma (NHL) activity of two novel CRM1 inhibitors (KPT-185 and KPT-276) in vitro and in vivo. KPT-185 displayed potent antiproliferative properties and induced cell-cycle arrest and apoptosis in several NHL cell ...

Crm1 is a mitotic effector of Ran-GTP in somatic cells

Nature Cell Biology, 2005
The Ran GTPase controls multiple cellular processes, including nuclear transport, mitotic checkpoints, spindle assembly and post-mitotic nuclear envelope reassembly. Here we examine the mitotic function of Crm1, the Ran-GTP-binding nuclear export receptor for leucine-rich cargo (bearing nuclear export sequence) and Snurportin-1 (ref. 3).

Nuclear Export as a Novel Therapeutic Target: The CRM1 Connection

Current Cancer Drug Targets, 2015
The integrity of eukaryotic cellular function depends on molecular and biochemical compartmentalization. The transport of macromolecules between compartments requires specific and energydriven mechanisms. It occurs through a class of transport proteins known as karyopherins, which are divided in three different groups (exportins, importins, and ...

CRM1 mediates ASC nuclear export and inflammasome activation

International Immunopharmacology
Inflammasomes are multiprotein complexes of the innate immune system that sense different pathogens or danger signals, and have been implicated in the pathogenesis of multiple human inflammatory diseases. The translocation of adaptor protein ASC from the nucleus to the cytosol is important for inflammasome assembly and activation, but the mechanism ...

The expression of CRM1 is associated with prognosis in human osteosarcoma.

Oncology reports, 2009
The nuclear export protein chromosomal region maintenance/exportin 1/Xpo1 (CRM1) is involved in the nuclear export of proteins and messenger RNAs and, thus, mediates the subcellular distribution of important molecules. Osteosarcoma is a ubiquitous and highly aggressive malignant bone tumor.
