Photocontrolled crRNA activation enables robust CRISPR-Cas12a diagnostics
CRISPR diagnostics based on nucleic acid amplification faces barriers to its commercial use, such as contamination risks and insufficient sensitivity. Here, we propose a robust solution involving optochemical control of CRISPR RNA (crRNA) activation in CRISPR detection.

Structural constraints and enzymatic promiscuity in the Cas6-dependent generation of crRNAs [PDF]
A hallmark of defense mechanisms based on clustered regularly interspaced short palindromic repeats (CRISPR) and associated sequences (Cas) are the crRNAs that guide these complexes in the destruction of invading DNA or RNA. Three separate CRISPR-Cas systems exist in the cyanobacterium Synechocystis sp. PCC 6803.

Programmable type III-A CRISPR-Cas DNA targeting modules. [PDF]
The CRISPR-Cas systems provide invader defense in a wide variety of prokaryotes, as well as technologies for many powerful applications. The Type III-A or Csm CRISPR-Cas system is one of the most widely distributed across prokaryotic phyla, and cleaves ...

PrimedSherlock: A tool for rapid design of highly specific CRISPR-Cas12 crRNAs
Abstract Background CRISPR-Cas based diagnostic assays provide a portable solution which bridges the benefits of qRT-PCR and serological assays in terms of portability, specificity and ease of use. CRISPR-Cas assays are rapidly fieldable, specific and have been rigorously validated against a number ...

Lactobacillus gasseri CRISPR-Cas9 characterization In Vitro reveals a flexible mode of protospacer-adjacent motif recognition. [PDF]
While the CRISPR-Cas9 system from S. pyogenes is a powerful genome engineering tool, additional programmed nucleases would enable added flexibility in targeting space and multiplexing. Here, we characterized a CRISPR-Cas9 system from L. gasseri and found

Stabilizing Functional Nucleic Acids for Real‐Sample Diagnostics
Functional nucleic acids enable programmable real‐sample diagnostics, but often lose function in complex biological matrices. This review highlights four stabilization levers—chemical, topological, architectural, and microenvironment/interface engineering—that preserve aptamer and DNAzyme performance in saliva, serum, urine, and stool, reframing ...

Kinetic dissection of pre-crRNA binding and processing by CRISPR-Cas12a
ABSTRACT CRISPR-Cas12a binds and processes a single pre-crRNA during maturation, providing a simple tool for genome editing applications. Here, we constructed a kinetic and thermodynamic framework for pre-crRNA processing by Cas12a in vitro , and we measured the contributions of ...

CRISPR-Cas12a nucleases function with structurally engineered crRNAs: SynThetic trAcrRNA
AbstractCRISPR-Cas12a systems are becoming an attractive genome editing tool for cell engineering due to their broader editing capabilities compared to CRISPR-Cas9 counterparts. As opposed to Cas9, the Cas12a endonucleases are characterized by a lack of trans-activating crRNA (tracrRNA), which reduces the complexity of the editing system and ...
