Cartoon model showing dynamic apo Lb2Cas12a and crRNA binding.

open access: yes, 2023
The conformational changes induced by crRNA binding are shown in this order: (1) Overall dynamic of apo Lb2Cas12a; (2) the process of crRNA binding; (3) stabilization of Lb2Cas12a-crRNA in compact conformation; (4) Met493~Leu523 undergoing major ...

Accessibility and Alignment of Trans‐Cleavage Substrates Modulate the Kinetics of Cas12a Ribonucleoprotein

open access: yesAngewandte Chemie, EarlyView.
Two key binding events that collectively determine the catalytic efficiency of trans‐cleavage: (1) the substrate passing through the gap between the REC and NUC domains, determined by Kpass, and (2) the alignment of the substrate with the catalytic triad of the ribonucleoprotein, determined by Kalign.

Synthetic Oligonucleotides Inhibit CRISPR-Cpf1-Mediated Genome Editing

open access: yesCell Reports, 2018
Summary: Previously, researchers discovered a series of anti-CRISPR proteins that inhibit CRISPR-Cas activity, such as Cas9 and Cpf1 (Cas12a). Herein, we constructed crRNA variants consisting of chemically modified DNA-crRNA and RNA-crRNA duplexes and ...

Efficient Multiple Genome Modifications Induced by the crRNAs, tracrRNA and Cas9 Protein Complex in Zebrafish. [PDF]

open access: yesPLoS ONE, 2015
The type II clustered regularly interspaced short palindromic repeats (CRISPR) associated with Cas9 endonuclease (CRISPR/Cas9) has become a powerful genetic tool for understanding the function of a gene of interest.

The Versatile Tool: CRISPR/Cas12a System for Nucleic Acid Detection

open access: yesShengwu huaxue yu shengwu wuli jinzhan
The CRISPR/Cas system consists of clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-associated genes (Cas). The system forms an adaptive immune system in archaea and bacteria.

Genetic Dissection of the Type III-A CRISPR-Cas System Csm Complex Reveals Roles of Individual Subunits

open access: yesCell Reports, 2019
Summary: The type III-A Csm complex of Streptococcus thermophilus (StCsm) provides immunity against invading nucleic acids through the coordinated action of three catalytic domains: RNase (Csm3), ssDNase (Cas10-HD), and cyclic oligoadenylates synthase ...

Highly specific chimeric DNA-RNA-guided genome editing with enhanced CRISPR-Cas12a system

open access: yesMolecular Therapy: Nucleic Acids, 2022
The clustered regularly interspaced short palindromic repeats (CRISPR)-Cas12a system is composed of a Cas12a effector that acts as a DNA-cleaving endonuclease and a crispr RNA (crRNA) that guides the effector to the target DNA.

Mutant p53R273H disrupts PDPK1 homodimerization and contributes to PDPK1 activation

open access: yesMolecular Oncology, EarlyView.
How mutant p53R273H drives AKT signaling is unclear. We show that p53R273H, but not wild‐type, directly binds PDPK1 via a mutation‐dependent conformational change. This interaction disrupts inhibitory PDPK1 homodimerization and enhances AKT phosphorylation.

Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

A CRISPR RNA Is Closely Related With the Size of the Cascade Nucleoprotein Complex

open access: yesFrontiers in Microbiology, 2019
The currently known prokaryotic adaptive immune system against mobile genetic elements is based on clustered regularly interspaced short palindromic repeats (CRISPR).
