Recent Advances in Gas Injection System-Free Cryo-FIB Lift-Out Transfer for Cryo-Electron Tomography of Multicellular Organisms and Tissues

open access: yesMicroscopy Today, 2022
Cryo-electron tomography (cryo-ET) enables visualization of protein complexes within their native cellular environment at molecular resolution. Most cells and all tissues, however, are too thick to be imaged directly by transmission electron microscopy ...

Cryo-focus ion beam (Cryo-FIB) and cryo-electron tomography (Cryo-ET) analyses of APCs in vps21Δ cells under conditions representative of nitrogen starvation.

open access: yes, 2022
Cells of vps21Δ were grown and treated as described in Fig 3A. After subjecting the cells to nitrogen starvation for 2 or 8 h, they were immediately analyzed using the Cryo-FIB and Cryo-ET methods, as described in the Materials and methods section.

A Modular Platform for Streamlining Automated Cryo-FIB Workflows [PDF]

open access: yes, 2021
Abstract Lamella micromachining by focused ion beam milling at cryogenic temperature (cryo-FIB) has matured into a preparation method widely used for cellular cryo-electron tomography. Due to the limited ablation rates of low Ga + ion beam currents required to maintain the ...

Heterogeneous non-canonical nucleosomes predominate in yeast cells in situ

open access: yeseLife, 2023
Nuclear processes depend on the organization of chromatin, whose basic units are cylinder-shaped complexes called nucleosomes. A subset of mammalian nucleosomes in situ (inside cells) resembles the canonical structure determined in vitro 25 years ago ...

Advanced cryo-tomography workflow developments - correlative microscopy, milling automation and cryo-lift-out [PDF]

open access: yes, 2020
Cryo-electron tomography (cryo-ET) is a groundbreaking technology for 3D visualisation and analysis of biomolecules in the context of cellular structures. It allows structural investigations of single proteins as well as their spatial arrangements within

Assembly and Imaging Set up of PIE-Scope

open access: yesBio-Protocol, 2020
Cryo-Electron Tomography (cryo-ET) is a method that enables resolving the structure of macromolecular complexes directly in the cellular environment. However, sample preparation for in situ Cryo-ET is labour-intensive and can require both cryo-lamella ...

Waffle Method: A general and flexible approach for improving throughput in FIB-milling

open access: yesNature Communications, 2022
Here the authors describe the Waffle Method, aimed at increasing the throughput of and solves several challenges present in cryo-FIB/SEM sample preparation for cryo-ET analysis — the highest-resolution method for obtaining 3D views of native biological ...
