Is mRNA decapping by ApaH like phosphatases present in eukaryotes beyond the Kinetoplastida?

open access: yesBMC Ecology and Evolution, 2021
Background ApaH like phosphatases (ALPHs) originate from the bacterial ApaH protein and have been identified in all eukaryotic super-groups. Only two of these proteins have been functionally characterised.

Tob1 is expressed in developing and adult gonads and is associated with the P-body marker, Dcp2 [PDF]

open access: yesCell and Tissue Research, 2015
Tob1 is a member of the BTG/TOB family of proteins with established antiproliferative function. In Danio rerio and Xenopus laevis, the Tob1 gene is expressed from the one-cell stage through to early gastrula stages, followed in later development by discrete expression in many tissues including the notochord and somites. In both mouse and human, Tob1 is

Investigating the regulation of the human decapping enzyme Dcp2 [PDF]

open access: yes, 2012
.Decapping is a critical step in several mRNA decay pathways that are important for the regulation of gene expression. A major decapping complex consists of the decapping enzyme Dcp2 as well as several decapping enhancers.

CSDE1 controls gene expression through the miRNA-mediated decay machinery

open access: yesLife Science Alliance, 2020
CSDE1 is a new component of miRNA-induced silencing complex and interacts with DCP1–DCP2 decapping machinery with potential roles in miRNA-mediated target degradation.

General decapping activators target different subsets of inefficiently translated mRNAs

open access: yeseLife, 2018
The Dcp1-Dcp2 decapping enzyme and the decapping activators Pat1, Dhh1, and Lsm1 regulate mRNA decapping, but their mechanistic integration is unknown. We analyzed the gene expression consequences of deleting PAT1, LSM1, or DHH1, or the DCP2 C-terminal ...

Phosphorylation of mRNA decapping protein Dcp1a by the ERK signaling pathway during early differentiation of 3T3-L1 preadipocytes.

open access: yesPLoS ONE, 2013
BackgroundTurnover of mRNA is a critical step in the regulation of gene expression, and an important step in mRNA decay is removal of the 5' cap. We previously demonstrated that the expression of some immediate early gene mRNAs is controlled by RNA ...

RNA decay is an antiviral defense in plants that is counteracted by viral RNA silencing suppressors. [PDF]

open access: yesPLoS Pathogens, 2018
Exonuclease-mediated RNA decay in plants is known to be involved primarily in endogenous RNA degradation, and several RNA decay components have been suggested to attenuate RNA silencing possibly through competing for RNA substrates.

Human Dcp2: a catalytically active mRNA decapping enzyme located in specific cytoplasmic structures [PDF]

open access: yesThe EMBO Journal, 2002
We have cloned cDNAs for the human homologues of the yeast Dcp1 and Dcp2 factors involved in the major (5'-3') and NMD mRNA decay pathways. While yeast Dcp1 has been reported to be the decapping enzyme, we show that recombinant human Dcp2 (hDcp2) is enzymatically active. Dcp2 activity appears evolutionarily conserved.

The ApaH-like phosphatase TbALPH1 is the major mRNA decapping enzyme of trypanosomes. [PDF]

open access: yesPLoS Pathogens, 2017
5'-3' decay is the major mRNA decay pathway in many eukaryotes, including trypanosomes. After deadenylation, mRNAs are decapped by the nudix hydrolase DCP2 of the decapping complex and finally degraded by the 5'-3' exoribonuclease. Uniquely, trypanosomes

Intrinsically disordered regions of Tristetraprolin and DCP2 directly interact to mediate decay of ARE-mRNA [PDF]

open access: yes, 2021
Abstract The RNA binding protein Tristetraprolin (TTP) is a potent activator of mRNA decay, specifically for transcripts bearing AU-rich elements (AREs) in their 3′-untranslated regions. TTP functions as a mediator for mRNA decay by interacting with the decay machinery and recruiting it to the target ARE-mRNA. In this
