The Mitochondrial Guardian α‐Amyrin Mitigates Alzheimer's Disease Pathology via Modulation of the DLK‐SARM1‐ULK1 Axis

open access: yesAdvanced Science, EarlyView.
Dietary habits play a key role in chronic diseases, and higher annual consumption of fruit and vegetable may lower risk of dementia. Artificial intelligence predicts the lipid‐like compound α‐Amyrin (αA) from plants with edible peels as a drug candidate against Alzheimer's disease.

HSP90 inhibitors stimulate DNAJB4 protein expression through a mechanism involving N6-methyladenosine. [PDF]

open access: yes, 2019
Small-molecule inhibitors for the 90-kDa heat shock protein (HSP90) have been extensively exploited in preclinical studies for the therapeutic interventions of human diseases accompanied with proteotoxic stress.

Histone Demethylases in Cancer [PDF]

open access: yesCurrent Pharmacology Reports, 2015
Epigenetic regulation, which involves with covalent modifications of DNA and the protein that bundles DNA, namely histones, is an exciting and variable phenomenon that affects normal cellular genetic character and contributes to human diseases. One key histone modification is methylation, regulated by methyl transferases and demethylases.

A Self‐Organized Liquid Reaction Container for Cellular Memory

open access: yesAdvanced Science, EarlyView.
How cells restore epigenetic information lost during replication is not known. This work proposes a mechanism based on the formation of biomolecular condensates. These condensates are induced by the chromosome itself and serve as reaction vessels for reconstructing missing epigenetic markers.

Histone H4K20 Demethylation by Two hHR23 Proteins

open access: yesCell Reports, 2020
Summary: Histone methyl groups can be removed by demethylases. Although LSD1 and JmjC domain-containing proteins have been identified as histone demethylases, enzymes for many histone methylation states or sites are still unknown.

KDM4A Erases the H3R17me2a Mark, Facilitating Chromosome Condensation

open access: yesAdvanced Science, EarlyView.
This study reveals a reversible histone modification switch governing chromosome condensation during mitosis. PKCα‐activated KDM4A removes H3R17me2a, permitting Suv39h1‐driven H3K9me3 deposition. This epigenetic transition recruits the chromosomal passenger complex and triggers Aurora B‐dependent H3S10 phosphorylation, coordinating chromatin remodeling

BGN/MDK Axis in the Melanoma Tumor Microenvironment Strengthens Tumor Malignancy by Modulating Cancer Cells and Cancer‐Associated Fibroblasts Crosstalk

open access: yesAdvanced Science, EarlyView.
This study reveals that m6A regulators cooperatively upregulate BGN in melanoma, promoting malignancy. Within the tumor microenvironment, CAFs show highest BGN expression. The BGN/MDK axis mediates cancer‐stroma crosstalk, driving normal fibroblast (NF) activation and enhancing the pro‐tumor effect of CAFs, highlighting a promising therapeutic target ...

JMJD1B Demethylates H4R3me2s and H3K9me2 to Facilitate Gene Expression for Development of Hematopoietic Stem and Progenitor Cells

open access: yesCell Reports, 2018
Summary: The arginine methylation status of histones dynamically changes during many cellular processes, including hematopoietic stem/progenitor cell (HSPC) development.

Data on molecular docking of tautomers and enantiomers of ATTAF-1 and ATTAF-2 selectivty to the human/fungal lanosterol-14α-demethylase

open access: yesData in Brief, 2020
The data have been obtained for tautomers and enantiomers of ATTAF-1 and ATTAF-2 that were developed based on antifungal standard drugs with triazole scaffold. These compounds were docked into the human and fungal lanosterol-14α-demethylase.
