USP32 regulates late endosomal transport and recycling through deubiquitylation of Rab7

open access: yesNature Communications, 2019
Though ubiquitin is known to broadly influence endosomal trafficking, few ubiquitin-utilizing enzymes targeting endosomal regulators are known. Here, the authors find that the deubiquitylating enzyme (DUB) USP32 influences endosomal membrane dynamics by ...

Fine-tuning the ubiquitin code at DNA double-strand breaks: deubiquitinating enzymes at work

open access: yesFrontiers in Genetics, 2015
Ubiquitination is a reversible protein modification broadly implicated in cellular functions. Signaling processes mediated by ubiquitin are crucial for the cellular response to DNA double-strand breaks (DSBs), one of the most dangerous types of DNA ...

T Cell Exhaustion in Cancer Immunotherapy: Heterogeneity, Mechanisms, and Therapeutic Opportunities

open access: yesAdvanced Science, EarlyView.
T cell exhaustion limits immunotherapy efficacy. This article delineates its progression from stem‐like to terminally exhausted states, governed by persistent antigen, transcription factors, epigenetics, and metabolism. It maps the exhaustion landscape in the TME and proposes integrated reversal strategies, providing a translational roadmap to overcome

Kinetic analysis of multistep USP7 mechanism shows critical role for target protein in activity

open access: yesNature Communications, 2019
Deubiquitinating enzymes (DUBs) are critical regulators of cellular processes by removing ubiquitin from specific targets. Here global kinetic modelling reveals the mechanism by which the low intrinsic activity of USP7 is substantially enhanced on a ...

Advances in Research on Ubiquitin Modification and Deubiquitinating Enzymes in Cancer Therapy [PDF]

open access: yesBIO Web of Conferences
Ubiquitination, as a classic protein post-translational modification, plays a key role in maintaining protein homeostasis. Protein ubiquitination is a constantly changing and reversible process.

TRIM40 Drives Pathological Cardiac Hypertrophy and Heart Failure via Ubiquitination of PKN2

open access: yesAdvanced Science, EarlyView.
This study identifies the E3 ligase TRIM40 as a key driver of pathological cardiac hypertrophy. TRIM40 binds PKN2 via its B‐box domain and, through its C29‐dependent catalytic activity, mediates K63‐linked ubiquitination of PKN2. This modification enhances PKN2 phosphorylation at Ser815, thereby driving hypertrophy.

Advances in Discovering Deubiquitinating Enzyme (DUB) Inhibitors

open access: yesJournal of Medicinal Chemistry, 2019
Deubiquitinating enzymes, or DUBs, comprise a family of proteases that regulate ubiquitination dynamics. Since their discovery, genetic and functional studies have nominated DUBs as a promising class for drug discovery across diverse therapeutic areas. Consequent probe and drug discovery efforts over the past 15 years have resulted in over 50 reported ...

Post‐Translational Regulation of CD8+ T Cell Fate and Dysfunction in Tumor Immunity

open access: yesAdvanced Science, EarlyView.
This review delineates how post‐translational modifications (PTMs) function as a central regulatory interface governing CD8+ T cell activation, differentiation, persistence, and exhaustion in antitumor immunity. By integrating antigenic, metabolic, and microenvironmental cues, diverse PTM programs coordinate transcriptional and chromatin states that ...

The role of deubiquitinating enzymes in spermatogenesis

open access: yesCellular and Molecular Life Sciences, 2015
Spermatogenesis is a complex process through which spermatogonial stem cells undergo mitosis, meiosis, and cell differentiation to generate mature spermatozoa. During this process, male germ cells experience several translational modifications. One of the major post-translational modifications in eukaryotes is the ubiquitination of proteins, which ...

USP5: Comprehensive insights into structure, function, biological and disease-related implications, and emerging therapeutic opportunities

open access: yesMolecular and Cellular Probes
Ubiquitin specific protease 5 (USP5) is a vital deubiquitinating enzyme that regulates various physiological functions by removing ubiquitin chains from target proteins. This review provides an overview of the structural and functional characteristics of
