DNA methylation profiling of the human major histocompatibility complex: A pilot study for the Human Epigenome Project [PDF]

open access: yes, 2004
The Human Epigenome Project aims to identify, catalogue, and interpret genome-wide DNA methylation phenomena. Occurring naturally on cytosine bases at cytosine-guanine dinucleotides, DNA methylation is intimately involved in diverse biological processes ...

A meta-analysis of epigenome-wide association studies in Alzheimer’s disease highlights novel differentially methylated loci across cortex

open access: yesNature Communications, 2021
Although epigenome-wide association studies of Alzheimer’s disease have highlighted neuropathology-associated DNA methylation differences, previous studies have been limited in sample size and brain region used.

Paternal Sperm Gnas‐ICR Epigenetic Programming Contributes to PPP‐Like Phenotypes in Female Offspring

open access: yesAdvanced Science, EarlyView.
A paternal glucocorticoid–sperm Gnas‐ICR axis is linked to ovarian endocrine programming in female offspring. Paternal preconception caffeine exposure elevates corticosterone, promotes sperm Gnas‐ICR hypermethylation, and is associated with ovarian Gnas upregulation, cAMP/PKA/CREB–StAR activation, enhanced estradiol synthesis, and PPP‐like phenotypes ...

Case report: Prenatal diagnosis of Kagami–Ogata syndrome in a Chinese family

open access: yesFrontiers in Genetics, 2022
The aim of this work was to explore the genetic cause of the proband (Ⅲ2) presenting with polyhydramnios and gastroschisis. Copy number variation sequencing (CNV-seq), methylation-specific multiplex PCR (MS-PCR), and methylation-specific multiplex ...

HMGCR‐Driven Cholesterol Metabolism Promotes Osteoarthritis Progression by Accelerating Synovial Fibroblast Senescence

open access: yesAdvanced Science, EarlyView.
In the pathological context of osteoarthritis (OA), the phosphorylation of AKT1 at Ser473 enhances its binding to Lys140 of Insig1, which facilitates the formation of AKT1–Insig1 complex. Subsequently, the activation of AKT1 promotes the phosphorylation of Insig1 at Ser189, potentially enhancing the dissociation of Insig1 from sterol regulatory element‑

TET1 Inhibition Promotes Therapeutic Sensitivity in TP53‐Mutant GBM by Influencing Genome Fragility and Altering TAMs Biology

open access: yesAdvanced Science, EarlyView.
In TP53mut GBM cells, reduced P53 function is associated with increased TET1 expression. Genetic or pharmacological inhibition of TET1 correlates with genome fragility, including DNA damage, cellular senescence, telomere shortening, and reactive oxygen species accumulation, which may contribute to increased efficacy of antitumor therapy.
