The Origin of Endoderm: Transmission Electron Microscopic Point of View
Objective: To show the transmission electron microscopic (TEM) evidence to confirm that the endoderm originates from the epiblast of the primitive streak or from other sources.

The tyrosine kinase Src is highly expressed in embryonic stem cells (ESCs) and ESC-differentiated cells, however, its functional role remains obscured. Here, we constitutively expressed Src in mouse ESCs and found these cells retained comparable levels ...

Distribution of tissue progenitors within the shield region of the zebrafish gastrula [PDF]
The zebrafish has emerged as an important model system for the experimental analysis of vertebrate development because it is amenable to genetic analysis and because its optical clarity allows the movements and the differentiation of individual cells to ...

Dissecting the dynamics of signaling events in the BMP, WNT, and NODAL cascade during self-organized fate patterning in human gastruloids. [PDF]
During gastrulation, the pluripotent epiblast self-organizes into the 3 germ layers-endoderm, mesoderm and ectoderm, which eventually form the entire embryo.

Signaling Centers Drive Brachyury Dynamics and Lineage Commitment in mESC Aggregates
We used a mouse embryonic stem cell aggregate model to compare the use of Chiron and BMP4 signaling as pulses (applied to the whole aggregate) or signaling centers (applied to part of the aggregate). BMP4 as a pulse was shown to drive neural differentiation, with Chiron inhibiting this effect.

Dynamics of Wnt activity on the acquisition of ectoderm potency in epiblast stem cells
During embryogenesis, the stringent regulation of Wnt activity is crucial for the morphogenesis of the head and brain. The loss of function of the Wnt inhibitor Dkk1 results in elevated Wnt activity, loss of ectoderm lineage attributes from the anterior ...

A critical aspect of chromosomal remodeling in small molecule‐induced multilineage trans‐differentiation(iMT) is elucidated. A dynamic atlas spanning over 35 days profiles terminal trans‐differentiated cells, which reverted to a more youthful state. This study reveals parallels between iMT and other cell fate determination processes, emphasizing an ...

In Vivo Differentiation Potential of Epiblast Stem Cells Revealed by Chimeric Embryo Formation
Chimera formation after blastocyst injection or morula aggregation is the principal functional assay of the developmental potential of mouse embryonic stem cells (ESCs).

Cell lineage specific distribution of H3K27 trimethylation accumulation in an in vitro model for human implantation. [PDF]
Female mammals inactivate one of their two X-chromosomes to compensate for the difference in gene-dosage with males that have just one X-chromosome. X-chromosome inactivation is initiated by the expression of the non-coding RNA Xist, which coats the X ...
