Candidiasis refers to both superficial and deep-tissue fungal infections often caused by Candida albicans. The treatment of choice for these infections is the use of azoles, such as fluconazole (FLC). However, the increased use of antifungal agents has led to the emergence of azole-resistant isolates of C. albicans. Thus, the development of alternative

Association of T916C (Y257H) mutation inCandida albicans ERG11with fluconazole resistance
Mycoses, 2012SummaryRepeated and prolonged use of fluconazole in treating candidosis leads to drug resistance. The aim of this study was to confirm the relationship between T916C (Y257H) mutation inCandida albicans ERG11and fluconazole resistance. We replaced one copy ofERG11withERG11containing the T916C mutation inC.

The ergosterol pathway in fungal pathogens is an attractive antimicrobial target because it is unique from the major sterol (cholesterol) producing pathway in humans. Lanosterol 14alpha-demethylase is the target for a major class of antifungals, the azoles. In this study we have isolated the gene for this enzyme from Cryptococcus neoformans.

Upregulation of the ERG11 gene in Candida krusei by azoles.
Daru : journal of Faculty of Pharmacy, Tehran University of Medical Sciences, 2012Candida species are the agents of local and systemic opportunistic infections and have become a major cause of morbidity and mortality in the last few decades. Azole resistance in Candida krusei (C. krusei) species appears to be the result of gene alterations in relation to the ergosterol biosynthesis pathway, as well as efflux pumps.

[Analysis of ERG11 gene mutation in Candida albicans].
Di 1 jun yi da xue xue bao = Academic journal of the first medical college of PLA, 2007To study the relationship between fluconazole (FCZ)-resistance of Candida albicans and the mutation of ERG11 gene encoding FCZ-targeted enzyme.Three strains of FCZ-susceptible and 10 FCZ-resistent C. albicans were isolated from the urethra, vagina, oropharynx, respiratory tract, prostate secretion and blood samples.

The mechanism of fluconazole resistance in Candida tropicalis is still unclear. Recently, we isolated a fluconazole-resistant strain of C. tropicalis from the blood specimen of a patient with candidemia in China. In vitro antifungal susceptibility of the isolate was determined by using CLSI M27-A3 and E-test methods. The sequence of ERG11 gene was then

ERG11 mutations and upregulation in clinical itraconazole-resistant isolates of Candida krusei
Canadian Journal of Microbiology, 2016To better understand the association between the ERG11 gene and drug resistance in Candida krusei, C. krusei strains were isolated from patients from January 2010 to May 2013. Susceptibility to 5-fluorocytosine (5-FC), amphotericin B (AMB), voriconazole (VRC), fluconazole (FLC), and itraconazole (ITR) were tested by broth microdilution method ...

Variation in the erg11 Gene from Pneumocystis jirovecii.
2010The erg11 gene codes for sterol 14-demethylase (14DM), a key enzyme in sterol biosynthesis and the target for triazole antimycotic drugs. Two sterol composition phenotypes have been described in patients with pneumonia caused by P. jirovecii; one phenotype has a much higher amount of lanosterol derivatives (C31 and C32 sterols) dominated by ...

The new mutation L321F in Candida albicans ERG11 gene may be associated with fluconazole resistance
Revista Iberoamericana de Micología, 2013For many years fluconazole has been commonly used to treat Candida infections. However, the indiscriminate use of this antimycotic therapy has favored the emergence of resistant isolates. Mutations in the ERG11 gene have been described as one of the primary mechanisms of resistance in Candida species. In this study we investigated missense mutations in ...
