Upregulation of the ERG11 gene in Candida krusei by azoles.

Daru : journal of Faculty of Pharmacy, Tehran University of Medical Sciences, 2012
Candida species are the agents of local and systemic opportunistic infections and have become a major cause of morbidity and mortality in the last few decades. Azole resistance in Candida krusei (C. krusei) species appears to be the result of gene alterations in relation to the ergosterol biosynthesis pathway, as well as efflux pumps.

Reduced susceptibility of Candida albicans clinical isolates to azoles and detection of mutations in the ERG11 gene

Diagnostic Microbiology and Infectious Disease, 2013
We investigated the susceptibility of Candida albicans isolated from clinic specimens to azole antifungal agents and estimated the association of the ERG11 mutations with azole resistance during recent 5years in China. In this study, novel mutations G346A, A434V, and L480F in ERG11 may be related to azole resistance in C. albicans.

Evaluation of the V404I and V509M amino acid substitutions of ERG11 gene in Candida albicans isolates by pyrosequencing

Folia Microbiologica, 2010
The molecular mechanisms underlying fluconazole resistance in C. albicans involve mutations and the overexpression of the ERG11 gene and membrane transport proteins. We examined the relationship between the reduced fluconazole susceptibility of C. albicans and mutations of V404I and V509M in the ERG11 gene in 182 C. albicans clinical isolates using the

Variation in the erg11 Gene from Pneumocystis jirovecii.

2010
The erg11 gene codes for sterol 14-demethylase (14DM), a key enzyme in sterol biosynthesis and the target for triazole antimycotic drugs. Two sterol composition phenotypes have been described in patients with pneumonia caused by P. jirovecii; one phenotype has a much higher amount of lanosterol derivatives (C31 and C32 sterols) dominated by ...

Expression of Lanosterol 14-Demethylase (ERG11) Gene of Three-Drug Combinations in Candida albicans

Iranian Journal of Pharmaceutical Sciences
Patients with impaired immunity are at particular risk of infections with Candida albicans. Antifungal drugs such as azoles commonly used for candidiasis treatment, but drug resistance is one of the most common problems for public health. The aim of this study was to evaluate the expression of lanosterol 14-demethylase (ERG11) gene for three-drug ...

Investigating role of positively selected genes and mutation sites of ERG11 in drug resistance of Candida albicans

Archives of Microbiology
The steep increase in acquired drug resistance in Candida isolates has posed a great challenge in the clinical management of candidiasis globally. Information of genes and codon sites that are positively selected during evolution can provide insights into the mechanisms driving antifungal resistance in Candida.

Investigation of ERG11 and MRR1 Resistance Genes in Candida parapsilosis Isolates

Candida parapsilosis is an important fungal pathogen that is particularly isolated from blood cultures. In recent years, although there are regional differences between centres, resistance to fluconazole which is the most commonly preferred drug for candidemia, has increased in many regions.

Cytochrome P450 lanosterol 14α-demethylase (CYP51): Insights from molecular genetic analysis of the ERG11 gene in Saccharomyces cerevisiae

The Journal of Steroid Biochemistry and Molecular Biology, 1992
Eukaryotes characteristically express a cytochrome P450-catalyzed sterol 14α-methyl demethylase as an essential step in the production of membrane sterols. Lanosterol 14α-demethylase of Saccharomyces cerevisiae is the best characterized representative of these enzymes among fungi and provides a model system for the molecular genetic analysis of the ...
