Integrated network pharmacology, molecular docking, and dynamics simulations revealed the anti‐osteoarthritis mechanism of Ermiao San. Key bioactive compounds (e.g., quercetin) synergistically target core proteins (e.g., TNF and IL‐6), modulating critical pathways (e.g., TNF signaling) to inhibit inflammation and cartilage degradation, demonstrating a ...

Recombinant expression and functional characterisation of regiospecific flavonoid glucosyltransferases from Hieracium pilosella L. [PDF]
Five glucosyltransferases were cloned by RT-PCR amplification using total RNA from Hieracium pilosella L. (Asteraceae) inflorescences as template. Expression was accomplished in Escherichia coli, and three of the HIS-tagged enzymes, UGT90A7, UGT95A1, and

Depigmenting Effects of Esculetin and Esculin Isolated from Fraxinus rhynchophyllaHance [PDF]
Stem bark extracts of Fraxinus rhynchophylla Hance were found to contain two major bioactive components, esculetin and esculin. Esculetin substantially inhibited melanogenesis in B16F10 melanoma cells, with an IC50 value of2.8 μm, and reduced melanin synthesis in Melan-A cells.

Study of Umbelliferone Hydroxylation to Esculetin Catalyzed by Polyphenol Oxidase
We characterize umbelliferone, a derivative of 2,4-dihydroxycoumaric acid, as a substrate of polyphenol oxidase. This enzyme hydroxylates umbelliferone to esculetin, its o-diphenol, and then oxidizes it to o-quinone. The findings show that umbelliferone, an intermediate in one of the coumarin biosynthesis pathways, may be transformed into its o ...
