FcRN receptor antagonists in the management of myasthenia gravis

open access: yesFrontiers in Neurology, 2023
Myasthenia gravis (MG) is an autoimmune disorder characterized by autoantibodies specifically directed against proteins located within the postsynaptic membrane of the neuromuscular junction.

IQ Survey Results on Current Industry Practices: Part 2—Quantitative Evaluations of Immunogenicity Assessment

open access: yesClinical Pharmacology &Therapeutics, EarlyView.
All biotherapeutics have the potential to induce an immunogenic response and generate anti‐drug antibodies (ADAs), especially when administered as multiple doses over prolonged periods. However, a clinically meaningful effect of ADAs can be difficult to identify to communicate the impact of immunogenicity on drug exposure, safety and efficacy outcomes ...

The Minimal Effect of Linker Length for Fatty Acid Conjugation to a Small Protein on the Serum Half-Life Extension

open access: yesBiomedicines, 2020
Conjugation of serum albumin or one of its ligands (such as fatty acid) has been an effective strategy to prolong the serum half-lives of drugs via neonatal Fc receptor (FcRn)–mediated recycling of albumin.

Integrative cross‐tissue analysis unveils complement‐immunoglobulin augmentation and dysbiosis‐related fatty acid metabolic remodeling during mammalian aging

open access: yesiMeta, EarlyView.
Our study provides a comprehensive multi‐omics profile of aging, integrating proteomic, metabolomic, and metagenomic analyses across diverse tissues and plasma. We identified the synergistic amplification of the circulating complement system and tissue‐wide immunoglobulin accumulation as key molecular drivers of inflammaging.

Efanesoctocog Alfa Population Pharmacokinetics and Repeated Time‐To‐Event Analysis of Bleeds in Adults, Adolescents, and Children with Severe Hemophilia A

open access: yesThe Journal of Clinical Pharmacology, EarlyView.
Abstract Efanesoctocog alfa is a first‐in‐class high‐sustained factor VIII (HSF) replacement therapy for treatment of hemophilia A. This article presents population pharmacokinetics (PopPK) of efanesoctocog alfa and repeated time‐to‐event (RTTE) analysis of bleeding episodes in adults/adolescents (≥12 years of age) and children (<12 years).

Histologic and molecular features shared between antibody‐mediated rejection of kidney allografts and chronic histiocytic intervillositis support common pathogenesis

open access: yesThe Journal of Pathology, EarlyView.
Abstract Chronic histiocytic intervillositis (CHI) is an inflammatory condition of the placenta, characterised by an abnormal, mainly macrophagic infiltrate within the intervillous space. Recent research suggests that CHI results from a ‘maternal‐foetal rejection’ mechanism, because at least some CHI cases fulfil the criteria for antibody‐mediated ...

Fully Convolutional Recurrent Network for Handwritten Chinese Text Recognition [PDF]

open access: yesarXiv, 2016
This paper proposes an end-to-end framework, namely fully convolutional recurrent network (FCRN) for handwritten Chinese text recognition (HCTR). Unlike traditional methods that rely heavily on segmentation, our FCRN is trained with online text data directly and learns to associate the pen-tip trajectory with a sequence of characters.

Harnessing Open‐Source Solutions: Insights From the First Open Systems Pharmacology (OSP) Community Conference

open access: yesCPT: Pharmacometrics &Systems Pharmacology, EarlyView.
ABSTRACT In 2017, the free and open‐source software Open Systems Pharmacology (OSP) was launched. Since then, OSP has evolved from a small community into a diverse network of stakeholders committed to advancing open‐source solutions for model‐informed drug development (MIDD). In this context, the first OSP Community Conference was hosted by Novartis in

Knockout of the neonatal Fc receptor alters immune complex trafficking and lysosomal function in cultured podocytes.

open access: yesPLoS ONE, 2023
Podocytes are key to preventing the filtration of serum proteins into the urine. Recent evidence also suggests that in immune mediated kidney diseases, podocytes are the targets of immune complexes (ICs).

Depth Estimation on Underwater Omni-directional Images Using a Deep Neural Network [PDF]

open access: yesarXiv, 2019
In this work, we exploit a depth estimation Fully Convolutional Residual Neural Network (FCRN) for in-air perspective images to estimate the depth of underwater perspective and omni-directional images. We train one conventional and one spherical FCRN for underwater perspective and omni-directional images, respectively.
