Measuring fast gene dynamics in single cells with time-lapse luminescence microscopy. [PDF]

open access: yes, 2014
Time-lapse fluorescence microscopy is an important tool for measuring in vivo gene dynamics in single cells. However, fluorescent proteins are limited by slow chromophore maturation times and the cellular autofluorescence or phototoxicity that arises ...

TEAD1 Enhances Exosome Secretion and Promotes Exosome‐Mediated Tissue Regeneration

open access: yesAdvanced Science, EarlyView.
TEAD1 functions as a crucial molecular switch regulating exosome secretion in various cell types. TEAD1 enhances exosome secretion by upregulating key proteins associated with exosome secretion, including RAB11, CD9, and SNAP23. This study reveals a novel role for TEAD1 in regulating exosome secretion and tissue regeneration, particularly in diabetic ...

β‐Triketone‐Based Ionizable Cationic Lipids Synthesized via Click Chemistry for siRNA Delivery

open access: yesAdvanced Science, EarlyView.
This study innovatively applies β‐triketone‐based click chemistry in the synthesis of ionizable cationic lipids (ICLs), which allows the generation of a lipid library in large quantities and in a short time. The ICLs identified achieved effective lipid nanoparticles‐mediated hepatic gene silencing in vivo using siRNA, and computational simulations are ...

GNL3 Orchestrates AR Transcriptional Programs to Drive Castration‐Resistant Prostate Cancer and Immune Evasion

open access: yesAdvanced Science, EarlyView.
GNL3 is a novel AR coregulator with dual coactivator and corepressor functions in prostate cancer (PCa). Our study uncovers a previously unrecognized mechanism by which the AR transcriptional complex integrates oncogenic signaling and immune suppression.

NIBAN2/FLII/RREB1 Axis Drives Glioma Stem Cell Malignancy via TLR3 Pathway Activation

open access: yesAdvanced Science, EarlyView.
NIBAN2, highly expressed in glioma stem‐like cells (GSCs), assembles with FLII and transcription factor RREB1 to form a nuclear complex. This complex transcriptionally activates stemness‐associated genes (e.g., CD44, NANOG) and metabolic enzymes (e.g., LDHA), thereby sustaining both transcriptional and metabolic stemness programs.

A higher spectral range of beetle bioluminescence with infraluciferin

open access: yesFrontiers in Bioengineering and Biotechnology, 2022
Coleopteran bioluminescence is unique in that beetle luciferases emit colors ranging between green (ca.550 nm) and red (ca.600 nm), including intermediate colors such as yellow and orange, allowing up to 3 simultaneous parameters to be resolved in vitro ...
