Humanized and Charge‐Optimized CSPG4‐Specific CAR‐T Cells show Enhanced Efficacy against Head and Neck Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
CSPG4 is identified as a high‐value, stemness‐associated target in HPV‐negative HNSCC. By implementing rational biophysical engineering, a humanized and charge‐optimized CAR is developed to overcome tonic signaling‐induced exhaustion. This strategy induces a profound transcriptomic shift toward a rejuvenated, stem‐like memory state, significantly ...

Near-infrared dual bioluminescence imaging in mouse models of cancer using infraluciferin

open access: yeseLife, 2019
Bioluminescence imaging (BLI) is ubiquitous in scientific research for the sensitive tracking of biological processes in small animal models. However, due to the attenuation of visible light by tissue, and the limited set of near-infrared bioluminescent ...

Stereoisomeric bio‐inversion key to biosynthesis of firefly d‐luciferin [PDF]

open access: yesFEBS Letters, 2006
The chirality of the luciferin substrate is critical to the luciferin–luciferase reaction producing bioluminescence. In firefly, the biosynthetic pathway of d‐luciferin is still unclear, although it can be synthesized in vitro from d‐cysteine. Here, we show that the firefly produces both d‐ and l‐luciferin, and that the amount of active d‐luciferin ...

Inhibition of SIRT7 Overcomes Radioresistance in Pancreatic Neuroendocrine Tumors by Reactivating MEN1 Expression

open access: yesAdvanced Science, EarlyView.
Pancreatic neuroendocrine tumors frequently silence MEN1 through epigenetic mechanisms. Here, SIRT7 recruits DNMT1 to the MEN1 promoter, drives hypermethylation, and enhances DNA repair. Inhibiting SIRT7 restores MEN1, reduces MRN complex abundance, impairs double‐strand break repair, and sensitizes PanNET models to radiation, supporting SIRT7 as a ...

ホタル生物発光における発光制御機構の化学生物学的研究 [PDF]

open access: yes, 2016
Firefly bioluminescence is an enzymatic reaction, which requires Mg2+, ATP, and O2. The mechanism of this bioluminescence is as follows; first, the luciferase catalyzes the adenylylation of the luciferin and then the oxygenation of the adenylylated ...

Hypoxia-inducible factor 1 alpha is required for the tumourigenic and aggressive phenotype associated with Rab25 expression in ovarian cancer [PDF]

open access: yes, 2016
The small GTPase Rab25 has been functionally linked to tumour progression and aggressiveness in ovarian cancer and promotes invasion in three-dimensional environments.

NUDT21 Drives T‐Cell Acute Lymphoblastic Leukemia Through Dual Regulation of Alternative Polyadenylation and Transcriptional Activation

open access: yesAdvanced Science, EarlyView.
In summary, our study defines a coordinated oncogenic model in which NUDT21 integrates alternative polyadenylation–dependent UBE2D3 stabilization and transcriptional activation to sustain MYC‐driven T‐ALL cell survival, thereby establishing NUDT21 as a central regulatory node and a promising therapeutic target.

Using an Adenosine Triphosphate Bioluminescent Assay to Determine Effective Antibiotic Combinations against Carbapenem-Resistant Gram Negative Bacteria within 24 Hours. [PDF]

open access: yesPLoS ONE, 2015
Current in vitro combination testing methods involve enumeration by bacterial plating, which is labor-intensive and time-consuming. Measurement of bioluminescence, released when bacterial adenosine triphosphate binds to firefly luciferin-luciferase, has ...

Protein sterilization method of firefly luciferase using reduced pressure and molecular sieves [PDF]

open access: yes, 1973
The sterilization of the protein fruitfly luciferase under conditions that prevent denaturation is examined. Denaturation is prevented by heating the protein in contact with molecular seives and under a reduced pressure of the order of 0.00005 ...
