CircSMEK1 Suppresses HCC via the hnRNPK‐IGF2‐AKT Axis: A Diagnostic Biomarker and Therapeutic Target

CircSMEK1 is downregulated in MASH/HCC and predicts poor prognosis. It suppresses tumor progression by promoting hnRNPK ubiquitination and inhibiting the IGF2/PI3K/AKT axis, while its loss activates immunosuppressive cancer‐associated fibroblasts. Tie2‐Dependent Mechanisms Influence Leptomeningeal Collateral Dynamics and Reperfusion Following Stroke

This work demonstrates that endothelial EphA4 limits Tie2 activation and curbs leptomeningeal collateral expansion after ischemic stroke. Pharmacological activation of Tie2 by an Ang‐1 mimetic or genetic loss of EphA4 stimulates collateral vessel growth, improves cerebral perfusion, and enhances neurological recovery.

Mechanical Cues Regulate Cargo Sorting and Export at the Golgi

A pathway is identified linking extracellular matrix stiffness to secretion via Src–FAK–AMPK–GBF1 signaling. Golgi‐localized GBF1 acts as a mechanotransducer whose phosphorylation controls post‐Golgi cargo sorting toward secretion or lysosomal degradation.

Interactions of Antibody Drug Conjugate Anti‐Tubulin and Topoisomerase I Inhibitor Payloads with Radiotherapy to Potentiate Immunotherapy

Antibody drug conjugates deliver their cytotoxic anti‐tubulin or topoisomerase I inhibitor payloads to tumors through cancer cell receptor targeting. The released drug payloads induce cellular changes that interact with radiotherapy resulting in radiosensitization that improves cancer cell kill and stimulates anti‐tumor immune responses.
