Cuprous Ions Binding to Cytosine and Guanine Oligonucleotides
Using a comprehensive set of nucleic acid‐related molecules, from individual nucleobases to oligonucleotides, we demonstrate the selective binding of Cu+ to cytosine‐containing oligonucleotides. Contrary to guanine‐containing oligonucleotides, cytosine‐rich oligonucleotides form a Cu+‐complex with a clear near‐UV signature and are able to stop redox ...

One‐electron oxidation of purine and pyrimidine bases of isolated and cellular DNA generates related reactive base radical cations that are converted into final decomposition products through deprotonation and hydration. In the case of guanine, the most susceptible nucleobase to one‐electron oxidants, 8‐hydroxy‐7,8‐dihydroguanyl radical, thus formed by

Folding landscape of a parallel G-quadruplex [PDF]
Circular dichroism and stopped-flow UV spectroscopies were used to investigate the thermodynamic stability and the folding pathway of d(TGAG3TG3TAG3TG3TA2) at 25C in solutions containing 25 mM KCl. Under these conditions the oligonucleotide adopts a thermally stable, all-parallel G-quadruplex topography containing three stacked quartets.

5‐Substituted 2‐(naphthalen‐2‐yl)‐4H‐chromen‐4‐ones are reported as a novel class of highly potent and selective triplex DNA ligands. These ligands induce triplex formation at submicromolar concentrations and inhibit enzymatic activity via ligand‐mediated triplex formation.

Harnessing the synergistic interplay of supramolecular self‐assembly, under macromolecular crowding conditions, and enzymatic‐mediated covalent crosslinking toward a stable protein‐based G‐quadruplex‐derived supramolecular bioink. This bioinspired strategy enables the biofabrication of complex and tunable ECM‐mimetic constructs, providing a platform ...

Supramolecular Assembly of Stepped‐Persistent ROS Nanogenerator for Sustained Tumor Immunotherapy
ROS catalytic systems often fail to achieve both effective tumor elimination and persistent immune activation due to their monotonous ROS production profiles. A stepped‐persistent ROS nanogenerator was developed by supramolecularly assembling multiple ROS generation modules within a nanofiber structure to simultaneously extend ROS generation duration ...

G‐Quadruplexes: Structural Diversity and Emerging Roles in Biomolecular Condensation
G‐quadruplexes (G4s) fold into diverse intra‐ and intermolecular structures, positioning them as emerging regulators of biomolecular condensation. Mechanistically, G4s autonomously form condensates, act as structural platforms to initiate and stimulate condensation, or induce phase transitions.

Relevance of G-quadruplex structures to pharmacogenetics
G-quadruplexes are non-canonical secondary structures formed within nucleic acids that are involved in modulating cellular processes such as replication, gene regulation, recombination and epigenetics.

Integrative in silico and in vitro screening of small molecules targeting RNA
Abstract Targeting structured RNA elements with small molecules has emerged as a promising yet technically challenging strategy for antiviral drug discovery. Here, we present a comprehensive and experimentally validated workflow for the integrative in silico and in vitro screening of RNA‐binding small molecules.

Mapping and characterization of G-quadruplexes in Mycobacterium tuberculosis gene promoter regions
Mycobacterium tuberculosis is the causative agent of tuberculosis (TB), one of the top 10 causes of death worldwide in 2015. The recent emergence of strains resistant to all current drugs urges the development of compounds with new mechanisms of action ...
