Molecular Dynamics‐Guided Sterol Engineering of mRNA‐Lipid Nanoparticles Reprograms Biodistribution and Enhances Spleen‐Selective Immunity

open access: yesAdvanced Science, EarlyView.
Sterol engineering enables precise control of mRNA lipid nanoparticles by tuning membrane organization. Replacing cholesterol with bile acid‐derived sterols redirects protein expression from the liver to the spleen. Notably, CA‐20 LNP enhances antigen‐specific immune responses while exhibiting a favorable safety profile, highlighting sterols as a ...

Ahcy Acts as an Effector of Hnf4a‐Driven Super‐Enhancer Activation to Alleviate MASLD During Intermittent Fasting

open access: yesAdvanced Science, EarlyView.
Our study identifies an Hnf4a‐driven super‐enhancer of hepatic Ahcy that mediates the protective effects of intermittent fasting against MASLD. Disruption of the Ahcy super‐enhancer reduces Ahcy expression and exacerbates lipid accumulation. This pathway prevents aberrant promoter hypermethylation by maintaining the SAM/SAH balance, as exemplified by ...

Datasets of microarray analysis to identify Gpr137b-dependent interleukin-4-responsive genes in the mouse macrophage cell line RAW264

open access: yesData in Brief, 2019
Macrophages are classified mainly into two subtypes, M1 and M2, which exhibit distinct phenotypes, based on their microenvironment. We have recently demonstrated that Gpr137b is abundantly expressed in RAW264 macrophages, “Gpr137b is an orphan G-protein ...

An experimental comparison of classification algorithms for hierarchical prediction of protein function [PDF]

open access: yes, 2007
Some data can be naturally organised as a hierarchy of classes. The classification of data in such a hierarchy poses some unique challenges to data miners. One particular case of this is the classification of GPCR proteins by their function.

Targeting Tex10 Overcomes Oxaliplatin Resistance by Competitively Disrupting the Non‐Canonical BAF Complex in Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
This study reveals that Tex10 drives oxaliplatin resistance in colorectal cancer by competitively binding BRD9 to disrupt the ncBAF complex, thereby suppressing AMBRA1 transcription and ULK1‐mediated autophagy. Gemcitabine is identified as a direct Tex10 inhibitor that restores autophagy and overcomes resistance.

Pharmacological Applications of Bile Acids and Their Derivatives in the Treatment of Metabolic Syndrome

open access: yesFrontiers in Pharmacology, 2018
Apart from well-known functions of bile acids in digestion and solubilization of lipophilic nutrients and drugs in the small intestine, the emerging evidence from the past two decades identified the role of bile acids as signaling, endocrine molecules ...

F7 Drives Gastric Cancer Metastasis Through Anoikis Resistance and Tumor Microenvironment Remodeling

open access: yesAdvanced Science, EarlyView.
ABSTRACT Coagulation factor VII (F7) has been implicated in tumor progression; however, its role in gastric cancer metastasis and immune evasion remains incompletely understood. In this study, we identified F7 as a clinically relevant driver of gastric cancer.

Die Funktion der Helix 8 für die Regulation des Bradykinin B2 Rezeptors [PDF]

open access: yes, 2012
Upon activation the human bradykinin B2 receptor (B2R) acts as guanine nucleotide exchange factor for the G proteins Gq/11 and Gi. Thereafter, it gets phosphorylated by G protein-coupled receptor kinases (GRKs) and recruits beta-arrestins, which block ...

Unravelling the therapeutic landscape of bile acid-based therapies in gastrointestinal disorders

open access: yesThe Saudi Journal of Gastroenterology
Bile acids serve as endogenous ligands for nuclear and cell membrane receptors and play a crucial role in bile acid and lipid metabolism. These detergent-like compounds promote bile flow and aid in the absorption of dietary fats and fat-soluble vitamins ...

Cholesterol 7α-hydroxylase protects the liver from inflammation and fibrosis by maintaining cholesterol homeostasis[S]

open access: yesJournal of Lipid Research, 2016
Cholesterol 7α-hydroxylase (CYP7A1) plays a critical role in control of bile acid and cholesterol homeostasis. Bile acids activate farnesoid X receptor (FXR) and Takeda G protein-coupled receptor 5 (TGR5) to regulate lipid, glucose, and energy metabolism.
