Unlocking the potential of pyroptosis in tumor immunotherapy: a new horizon in cancer treatment
BackgroundThe interaction between pyroptosis—a form of programmed cell death—and tumor immunity represents a burgeoning field of interest. Pyroptosis exhibits a dual role in cancer: it can both promote tumor development and counteract it by activating ...

Caspase-1 activates gasdermin A in non-mammals
Gasdermins oligomerize to form pores in the cell membrane, causing regulated lytic cell death called pyroptosis. Mammals encode five gasdermins that can trigger pyroptosis: GSDMA, B, C, D, and E.

Monitoring gasdermin pore formation in vitro
The gasdermin (GSDM) family consists of gasdermin A (GSDMA), B (GSDMB), C (GSDMC), D (GSDMD), E or DNFA5 (GSDME), and DFNB59 in human. Expressed in the skin, gastrointestinal tract, and various immune cells, GSDMs mediate homeostasis and inflammation upon activation by caspases and unknown proteases.

Role of RIPK1/RIPK3/MLKL signalling pathway in sepsis‐associated acute kidney injury
Abstract Sepsis‐associated acute kidney injury (SA‐AKI) is a common clinical syndrome in critically ill patients, and its high mortality rate is closely related to complex pathological mechanisms. Existing studies have shown that the pathophysiological process of SA‐AKI involves complex multi‐mechanism interactions, including an uncontrolled systemic ...

ABSTRACT Acute kidney injury (AKI) is a global health problem with significant long‐term harm if the prognosis is poor. Dual‐specificity phosphatase 2 (DUSP2) is involved in key regulatory pathways in several disease processes, but its function in renal pathophysiology is unclear.

Abstract Type 2 diabetes mellitus (T2DM) significantly increases the risk and severity of cerebral ischaemia–reperfusion (IR) injury, yet effective neuroprotective treatments for diabetic stroke remain limited. This study explored whether a combination of sinomenine and metformin could offer enhanced neuroprotection in diabetic rats subjected to ...

Inhibiting separase with the specific inhibitor Sepin‐1 has been found to effectively enhance gefitinib‐induced cytotoxic in lung cancer cell. Mechanically, separase inhibitor Sepin‐1 plus gefitinib decreases the expression of PTBP1 and TAK1 to trigger RIPK1‐dependent PANoptosis. ABSTRACT Gefitinib is the most frequently employed anti‐lung cancer drug,
