Functions of MDA5 and its domains in response to GCRV or bacterial PAMPs

Fish & Shellfish Immunology, 2015
Melanoma differentiation-associated gene 5 (MDA5) is a member of retinoic acid-inducible gene I (RIG-I)-like receptor (RLR) family which can initiate type I IFN expression in response to RNA virus infection. In this study, we constructed six mutants of Ctenopharyngodon idella MDA5 (CiMAD5) overexpression plasmids and generated stable transfected C ...

Cloning of six serpin genes and their responses to GCRV infection in grass carp (Ctenopharyngodon idella)

open access: yesFish and Shellfish Immunology, 2019
Grass carp, an economically important aquaculture fish, is very sensitive to Grass Carp Reovirus (GCRV). Haemorrhagic disease caused by GCRV infection can cause large-scale death of first-year grass carp, thereby severely restricting the intensive ...

bcIRF5 activates bcTBK1 phosphorylation to enhance PANoptosis during GCRV infection

Fish and Shellfish Immunology
TBK1 is an important IFN antiviral signalling factor, and in previous work black carp TBK1 (bcTBK1) and black carp IRF5 (bcIRF5) together promoted cell death in GCRV-infected cells. In this research, bcTBK1 and bcIRF5 were investigated both in vivo and in vitro to delineate their individual and combined functions.

ANAX4 is a downstream molecule of LGP2 and promotes GCRV proliferation

Fish & Shellfish Immunology
This study explored the key molecules and signal pathways in the pathogenesis of grass carp reovirus (GCRV). Using immunoprecipitation mass spectrometry and Co-IP validation, the protein CiANXA4 was identified which interacts indirectly with CiLGP2. CiANXA4 encodes 321 amino acids, including 4 ANX domains.

Enhanced Expression of GCRV VP6 in CIK Cells by Relative Sequence Optimization

Applied Biochemistry and Biotechnology, 2014
Efficient expression of target protein is one of strategies for gene therapy or vaccine design. Many studies showed that codon optimization could enhance the expression of target proteins. In this paper, a target sequence of about 1.26 kb encoding the major capsid protein VP6 of grass carp reovirus (GCRV) and an optimized counterpart were synthesized ...

Baculovirus-mediated GCRV vp7 and vp6 genes expression in silkworm and grass carp

Molecular Biology Reports, 2016
Grass carp hemorrhagic disease is a common fish disease and often results in significant economic losses in grass carp aquaculture in China. This study was aimed to develop a novel oral vaccine against grass carp reovirus (GCRV). GCRV vp6 and vp7 genes with β-actin promoter of Megalobrama amblycephala and polyhedrin promoter (Ph10) of baculovirus ...

miR-2188-5p promotes GCRV replication by the targeted degradation of klf2a in Ctenopharyngodon idellus

Developmental & Comparative Immunology, 2023
Studies on host immunity evasion by aquatic viruses have largely focused on coding genes. There is accumulating evidence for the important biological functions of non-coding miRNAs in virus-host interactions. The regulatory functions of non-coding miRNAs in fish reovirus-host interactions remain unknown.

RBX1 and RBX2 promote GCRV replication in grass carp (Ctenopharyngodon idella)

Fish & Shellfish Immunology
Cullin-RING ligases (CRLs) constitute the most diverse E3 ligase family in the ubiquitin-proteasome system, yet their antiviral roles in bony fish remain poorly understood. Here, we identify the catalytic cores of CRLs, the RING-box proteins RBX1 and RBX2, as key modulators of antiviral signaling in cyprinid fish.

Ctenopharyngodon idellus DDX41 initiates IFN I and ISG15 expression in response to GCRV infection

Fish & Shellfish Immunology, 2020
As a member of DExD/H-box helicase family, DDX41 (DEAD box polypeptide 41) acts as an intracellular DNA sensor that induces type I IFN expression in mammals. Fish DDX41 shares some similar properties with the mammalian counterparts. In this study, a DDX41 orthologous gene from grass carp (Ctenopharyngodon idellus) (CiDDX41) was cloned and characterized.

IFNa of triploid hybrid of gold fish and allotetraploid is an antiviral cytokine against SVCV and GCRV

Fish & Shellfish Immunology, 2016
Triploid hybrid of gold fish (Carassius auratus red var., ♀, 2n = 100) and allotetroploid (♂, 2n = 100) displays much improved disease resistance than its parents. In this paper, one of the type I IFNs of triploid hybrid (3nIFNa) has been cloned and characterized.
