Gene Cloning and Recombinant DNA Libraries
2023Gene cloning involves the replication of DNA fragments in host organisms to produce identical copies. It serves as a vital tool for the production of proteins, vaccines, and therapeutic treatments. On the other hand, rDNA libraries—systematic collections of cloned DNA sequences—function as extensive genomic and cDNA archives.

Epitope Mapping with Gene Fragment Libraries
2001Linear epitopes, recognized by a large number of antibodies, are easily identified by expression of small fragments of the known antigen at the tip of filamentous phage. Immobilized antibodies, incubated with such a library of peptide-displaying phage, bind selectively to the fragments containing the epitope, therefore immobilizing the peptide-phage ...

Molekuliarnaia biologiia, 1990
Main approaches and methods used for constructing "jamping" and cDNA libraries, including novel ones, that omit the employment of methylases and linkers, are presented. The advantages and drawbacks of the well-known and new lambda vectors, suitable for the purposes mentioned, are discussed.

Cloning Genes From an Inner Ear cDNA Library
Archives of Otolaryngology - Head and Neck Surgery, 1993A rat inner ear complementary DNA (cDNA) library containing 1.9 x 10(6) recombinants was constructed and evaluated. Inserts averaged 2.0 (+/- 2.1) kilobases in length. A subset of inserts was screened for site of expression. Two cDNA transcripts were isolated based on cochlear expression restricted to the spiral ganglion.

Gene Libraries: Coverage, Efficiency and Diversity
2006Gene libraries are a biological mechanism for generating combinatorial diversity in the immune system. However, they also bias the antibody creation process, so that they can be viewed as a way of guiding lifetime learning mechanisms. In this paper we examine the implications of this view, by examining coverage, avoidance of self, clustering and ...

Construction of gene libraries for each human chromosome
Cytometry, 1990AbstractWe describe the construction of two complete sets of small insert, complete digest DNA libraries for each of the 24 human chromosomal types by the National Laboratory Gene Library Project. Flow sorting was used to purify the chromosomes which provided the DNA for cloning.

Selection of Recombinant Antibodies From Antibody Gene Libraries
2007After the sequencing of the human genome is completed, the research focus shifts toward the analysis of gene products. The human genome encodes more than 30,000 genes. Owing to alternative mRNA splicing and posttranslational modifications, for example, glycosylation, phoshorylation, and so on, the number of different proteins of human proteome is ...

Rational design of orthogonal libraries of protein coding genes
Proceedings of the ACM Conference on Bioinformatics, Computational Biology and Biomedicine, 2012Array-based oligonucleotide synthesis technologies provide access to thousands of custom-designed sequence variants at low cost. Large-scale synthesis and high-throughput assays have become valuable experimental tools to study in detail the interplay between sequence and function.

[The library of Rickettsia prowazekii genes].
Molekuliarnaia genetika, mikrobiologiia i virusologiia, 1987The DNA of Rickettsia provazekii strain E was cleaved by PstI restriction endonuclease under the conditions of partial restriction. The fragments were inserted into the PstI site of pBR325 and cloned in this plasmid. E. coli strain HB101 was used as a recipient for cloning.

Selection of Recombinant Antibodies from Antibody Gene Libraries
2013Antibodies are indispensable detection reagents for research and diagnostics and represent the biggest class of biological therapeutics on the market. In vitro antibody selection systems offer many advantages over animal-based technologies because the whole selection process is independent of the in vivo immune response.
