Functional Analysis of the vpx, vpr, and nef Genes of Simian Immunodeficiency Virus
Journal of Acquired Immune Deficiency Syndromes & Human Retrovirology, 1995The role of the vpx, vpr, and nef genes in the replication of simian immunodeficiency virus (SIV) was investigated using point and deletion mutations in these genes. The effects on replication kinetics of single or combined mutants--vpx, vpr, vpx-vpr, vpx-nef, vpr-nef, and vpx-vpr-nef--in established lymphoid CEMx174 and MT-4 cells were negligible ...

Importance of the nef gene for maintenance of high virus loads and for development of AIDS
Cell, 1991When rhesus monkeys were infected with a form of cloned SIVmac239 having a premature stop signal at the 93rd codon of nef, revertants with a coding codon at this position quickly and universally came to predominate in the infected animals. This suggests that there are strong selective forces for open functional forms of nef in vivo.

The HIV-1 nef gene acts as a positive viral infectivity factor
Trends in Microbiology, 1994The gene product of the nef gene of HIV-1 acts both to increase the infectivity of viral particles and to reduce the expression of the CD4 receptor molecule on the cell surface. These two functions of Nef may be related, in that downregulation of CD4 may promote the production of HIV that has greater infectivity.

One explanation of Nef gene behavior
Medical Hypotheses, 1999The Nef (negative factor) gene of primate retroviruses may serve an important evolutionary function. Selection pressures in the natural world, may at times, demand that retroviral infection not cause disease in a newly entered host species. Genetic alterations in the Nef gene may function to permit retroviral speciation by lowering retroviral ...

Biochemical and biological comparison of HIV-1 NEF and ras gene products
Virology, 1991Human immunodeficiency virus type 1 (HIV-1) NEF protein has been reported to share certain biochemical and structural properties with known oncoproteins like src or rats. To determine whether this is a general property of NEF from various HIV isolates, three different NEF proteins were expressed in Escherichia coli using a thermoinducible expression ...

Minor nef gene alterations after human HIV-DNA immunisation
AIDS, 2004Nef gene sequences were analysed in HIV-1-infected patients, immunised with a single nef gene or a combination of nef, rev and tat genes. After three to six immunisations, we found no increase of amino acid substitutions within predicted cytotoxic T-cell epitopes, compared with epitopes located outside predicted sites.

The nef Gene from a Long-Term HIV Type 1 Nonprogressor
AIDS Research and Human Retroviruses, 1996We examined the nef gene of HIV-1 in a long-term nonprogressor to look for evidence suggesting an attenuated virus. The nef gene was previously shown to be required for induction of AIDS. Simian immunodeficiency virus (SIV) deleted in nef, while infectious, fails to sustain the high viral loads necessary for the induction of AIDS in infected adult ...

Protective Effects of a Live Attenuated SIV Vaccine with a Deletion in the nef Gene
Science, 1992Vaccine protection against the human immunodeficiency virus (HIV) and the related simian immunodeficiency virus (SIV) in animal models is proving to be a difficult task. The difficulty is due in large part to the persistent, unrelenting nature of HIV and SIV infection once infection is initiated.

The HIV-1 nef gene product is associated with phosphorylation of a 46 kD cellular protein
AIDS, 1992To study the biochemical properties of the HIV-1 nef gene product.Earlier reports suggested that Nef protein is phosphorylated and has kinase activity. These properties were examined using an in vitro translated product.A DNA fragment encoding Nef of HIV-1SF2 was transcribed in vitro under the control of T7 promoter.

Intrahost and Interhost Variability of the HIV Type 1 nef Gene in Brazilian Children
AIDS Research and Human Retroviruses, 2009Many aspects of HIV-1 pathogenesis are affected by Nef protein activity, and efforts have been made to study variation in the nef gene and how that variation relates to disease outcome. We studied the genetic diversity of the nef gene in distinct clones obtained from the same patient (intrahost) and in sequences obtained from different hosts (interhost)
