Creb3l1 Overexpression Improves Flexor Tendon Repair in Aged Rats

open access: yesAging Cell, Volume 25, Issue 9, September 2026.
Aging impairs flexor tendon repair, with aged tendons showing reduced Col1a1 and Sparc expression and compromised mechanical properties. The transcription factor Creb3l1 binds both gene promoters and drives their expression; its overexpression restores Col1a1 and Sparc levels and improves the gliding function, healing strength, and elastic modulus of ...

Aging‐Related Myocardial Susceptibility Lowers Cardiac Tolerance to Chronic Intermittent Hypoxia Through Dynamin‐Related Protein 1‐Associated Mitochondrial Vulnerability

open access: yesAging Cell, Volume 25, Issue 9, September 2026.
Chronic intermittent hypoxia (CIH) induces early myocardial remodeling and senescence‐associated injury without overt systolic failure in young mice. Aging‐related susceptibility, modeled in vivo by Tert−/− mice and in vitro by D‐gal‐treated primary cardiomyocytes, amplifies CIH‐associated cardiac dysfunction and cellular injury.

Piezo1 Activation Rescues Anabolic Response to Mechanical Loading in Aged Bone via Connexin 43 Hemichannels

open access: yesAging Cell, Volume 25, Issue 9, September 2026.
Yoda1‐induced Piezo1 activation enhances Piezo1‐Cx43 interaction upon mechanical stimulation, triggering PI3K‐Akt signaling to drive Cx43 phosphorylation and hemichannel opening. These events elevate PGE2 release, suppress SOST expression in senescent osteocytes, and ultimately enhance bone anabolism on the endosteal surface.

DHCR24 Alleviates DNA Damage in Senescent Vascular Endothelial Cells via ENKUR/Ca2+ Signaling

open access: yesAging Cell, Volume 25, Issue 9, September 2026.
In senescent vascular endothelial cells, DHCR24 deficiency triggers ENKUR‐dependent Ca2+ overload, which in turn induces endoplasmic reticulum stress and mitochondrial dysfunction, ultimately exacerbating oxidative stress and DNA damage. ABSTRACT DNA damage is considered one of the major contributors to aging.

SIRT1 Silences L1 Retrotransposons by Stabilizing Heterochromatin‐Modifying Complexes

open access: yesAging Cell, Volume 25, Issue 9, September 2026.
SIRT1 binds the LINE‐1 5′‐UTR and recruits the heterochromatin modifiers Lamin B1 and KAP1 to maintain H3K9me3 levels, thereby suppressing LINE‐1 retrotransposition. SIRT1 deficiency enhances LINE‐1 transcription, leading to upregulation of senescence‐associated secretory phenotype factors, activation of the cGAS‐STING pathway, and consequent cellular ...
