Phenome-wide association study on miRNA-related sequence variants: the UK Biobank

open access: yesHuman Genomics, 2023
Background Genetic variants in the coding region could directly affect the structure and expression levels of genes and proteins. However, the importance of variants in the non-coding region, such as microRNAs (miRNAs), remain to be elucidated.

Analysis of a Population Genetics Model with Mutation, Selection, and Pleiotropy [PDF]

open access: yesJournal of Statistical Physics, 1999
We investigate the behavior of a population genetics model introduced by Waxman and Peck incorporating mutation, selection, and pleiotropy. The population is infinite and continuous variation of genotype is allowed. Nonetheless, Waxman and Peck showed that if the degree of pleiotropy is large enough,in this model a nonzero fraction of the population ...

Estimating quantitative genetic parameters in wild populations:A comparison of pedigree and genomic approaches [PDF]

open access: yes, 2014
The estimation of quantitative genetic parameters in wild populations is generally limited by the accuracy and completeness of the available pedigree information. Using relatedness at genome-wide markers can potentially remove this limitation and lead to

Investigating the effect of sexual behaviour on oropharyngeal cancer risk: a methodological assessment of Mendelian randomization

open access: yesBMC Medicine, 2022
Background Human papilloma virus infection is known to influence oropharyngeal cancer (OPC) risk, likely via sexual transmission. However, sexual behaviour has been correlated with other risk factors including smoking and alcohol, meaning independent ...

Statistical methods to detect pleiotropy in human complex traits [PDF]

open access: yesOpen Biology, 2017
In recent years pleiotropy, the phenomenon of one genetic locus influencing several traits, has become a widely researched field in human genetics. With the increasing availability of genome-wide association study summary statistics, as well as the ...

Rheumatoid arthritis and osteoporosis: shared genetic effect, pleiotropy and causality

open access: yesHuman Molecular Genetics, 2021
Abstract Rheumatoid arthritis (RA) is associated with increased localized and generalized bone loss, but the complex genetic mechanism between them is still unknown. By leveraging large-scale genome-wide association studies summary statistics and individual-level datasets (i.e.

Antagonistic pleiotropy and genetic polymorphism: a perspective [PDF]

open access: yesHeredity, 1999
Antagonistic pleiotropy, in which reproduction and viability counter each other, appears to be widely thought of great significance in life history theory and the evolution of senescence. However, the conditions for maintenance of polymorphism by antagonistic pleiotropy are quite restrictive.

Chronobiology of Cancer: How Aging Fuels Oncogenesis at the Molecular Level

open access: yesAging and Cancer, EarlyView.
This graphical abstract illustrates the key biological pathways linking aging with cancer development and progression. In the upper left, cumulative exposure to ultraviolet radiation, toxins, and reactive oxygen species (ROS) causes DNA damage and genomic instability, whereas age‐related decline in repair mechanisms, such as ATM/ATR, BER, and NER ...

A novel framework with automated horizontal pleiotropy adjustment in mendelian randomization

open access: yesHGG Advances
Summary: The presence of horizontal pleiotropy in Mendelian randomization (MR) analysis has long been a concern due to its potential to induce substantial bias.

MULTIPLE EQUILIBRIA AND MAINTENANCE OF ADDITIVE GENETIC VARIANCE IN A MODEL OF PLEIOTROPY [PDF]

open access: yesEvolution, 1990
We describe a multilocus model that incorporates pleiotropic stabilizing selection on a large number of characters. We find many different stable equilibria with different levels of polymorphism and additive genetic variability. The results lend support to Wright's concept of a complex adaptive surface with many peaks of different heights.
