Elevated exogenous palmitic acid promotes the S‐palmitoylation of SRPK1 in endothelial cells, a dynamic process governed by ZDHHC24 and APT1. This post‐translational modification strengthens the interaction between SRPK1 and the E3 ubiquitin ligase MIB1, thereby facilitating the proteasomal degradation of SRPK1.

NFE2L2 and SLC25A39 drive cuproptosis resistance through GSH metabolism
Cuproptosis is a recently discovered form of regulated cell death triggered by mitochondrial copper accumulation and proteotoxic stress. Here, we provide the first evidence that glutathione (GSH), a major non-protein thiol in cells, acts as a cuproptosis

Identification of an important cysteine residue in human glutamate–cysteine ligase catalytic subunit by site-directed mutagenesis [PDF]
Glutamate–cysteine ligase (GLCL) catalyses the rate-limiting step in glutathione biosynthesis. To identify cysteine residues in GLCL that are involved in its activity, eight conserved cysteine residues in human GLCL catalytic subunit (hGLCLC) were replaced with glycine residues by PCR-based site-directed mutagenesis.

This study identifies Pirin (PIR), an iron‐binding protein, as a critical ferroptosis suppressor in colorectal cancer through lipid membrane remodeling. PIR, induced by NRF2 during ferroptotic stress, transcriptionally regulates PLA2G4A to shift cellular lipid composition away from ferroptosis‐permissive polyunsaturated phospholipids.

Differential regulation of glutamate–cysteine ligase subunit expression and increased holoenzyme formation in response to cysteine deprivation [PDF]
GCL (glutamate–cysteine ligase) is a heterodimer of a GCLC (GCL catalytic subunit) that possesses all of the enzymatic activity and a GCLM (GCL modifier subunit) that alters the Ki of GCLC for GSH. We hypothesized that the expression of GCLM and the association of GCLM with GCLC were responsible for the apparent increase in GCL activity state observed ...

Staphylococcus aureus (S. aureus) infection creates a high‐lactate microenvironment, promoting p300‐mediated lactylation of PGK1 at lysine 361 (K361). Lactylated PGK1 translocates to the mitochondrial outer membrane and interacts with VDAC3. This interaction triggers FtMt downregulation, iron accumulation, and excessive PINK1/Parkin‐mediated mitophagy,

Cysteine Is the Metabolic Signal Responsible for Dietary Regulation of Hepatic Cysteine Dioxygenase and Glutamate Cysteine Ligase in Intact Rats [PDF]
Cysteine, rather than a precursor or metabolite of cysteine, appears to mediate the upregulation of cysteine dioxygenase (CDO) and the downregulation of glutamate cysteine ligase (GCL) in cultured primary rat hepatocytes. However, similar experiments in intact rats have not been performed to confirm in vivo that changes in hepatic cysteine levels are ...

The interaction between mitofusin 2 (MFN2) and phosphofurin acidic cluster sorting protein 2 regulates the formation of mitochondria‐associated endoplasmic reticulum membranes (MAM). Overexpression of MFN2 can promote the formation of MAMs and inhibit inflammation and oxidative stress response in mammary epithelial cells.
