Integrated PANoptosis Profiling Identifies Immunosuppressive Subtypes and a Prognostic Signature With Functional Validation of MLKL in Glioblastoma

open access: yesAnnals of Clinical and Translational Neurology, EarlyView.
ABSTRACT Objective The prognosis of glioblastoma (GBM) remains highly unfavorable, largely due to high tumor heterogeneity and an immunosuppressive microenvironment. However, the functional role of PANoptosis in this context is poorly understood. Methods Patients were stratified via K‐means clustering. A risk score model was constructed using prognosis‐

Reprogramming the Immunosuppressive Microenvironment in Triple‐Negative Breast Cancer via FAP‐Targeted Nanoprobes for Multimodal Imaging and Photothermal Therapy

open access: yesAdvanced Materials, EarlyView.
Targeting cancer‐associated fibroblasts (CAFs) with fibroblast activation protein (FAP)‐directed nanoprobes for multimodal imaging and photothermal remodeling of the immunosuppressive microenvironment to enhance immunotherapy in triple‐negative breast cancer (TNBC).

Stromal‐Derived IL‐8 Promotes M2 Macrophage Polarization via the RhoA/MRTF‐A/SRF Transcriptional Axis to Impair CD8+ T Cell Cytotoxicity in Lung Cancer

open access: yesAdvanced Science, EarlyView.
In the recondite pulmonary neoplastic milieu, DESMIN+ cancer‐associated fibroblasts function as the paramount stromal source of IL‐8, precipitating a deleterious immunological metamorphosis by engaging CXCR2 receptors on the myeloid lineage. This paracrine stimulus triggers a mechanotransduction cascade characterized by RhoA‐dependent actin ...

Genetic variants of human granzyme B predict transplant outcomes after HLA matched unrelated bone marrow transplantation for myeloid malignancies.

open access: yesPLoS ONE, 2011
Serine protease granzyme B plays important roles in infections, autoimmunity, transplant rejection, and antitumor immunity. A triple-mutated granzyme B variant that encodes three amino substitutions (Q48R, P88A, and Y245H) has been reported to have ...

RNA Helicase DDX21 Controls CD4+ T Cell Proliferation and Promotes Inflammatory Bowel Disease via Translational Control

open access: yesAdvanced Science, EarlyView.
ABSTRACT Inflammatory bowel disease (IBD) is characterized by dysregulated T cell responses. RNA helicases, including DExD‐box helicase 21 (DDX21), are pivotal in RNA metabolism, but their role in T cell‐mediated pathology during IBD remains unclear. Here, we demonstrate that DDX21 expression in CD4+ T cells correlates with cell cycle and translation ...

A 3D Human Bone and Bone Marrow‐on‐a‐Chip Model for In Vitro Bone Remodeling and Immune Cell Maintenance

open access: yesAdvanced Science, EarlyView.
This study presents a human Bone (and Bone Marrow)‐on‐a‐Chip model based on native human bone scaffolds and autologous cells. Dynamic perfusion and sequential cell seeding replicate the physiological bone remodeling process in vitro, enabling the long‐term culture of functional, mature bone marrow immune subpopulations.

Generation of an artificial human B cell line test system using Transpo-mAbTM technology to evaluate the therapeutic efficacy of novel antigen-specific fusion proteins. [PDF]

open access: yesPLoS ONE, 2017
The antigen-specific targeting of autoreactive B cells via their unique B cell receptors (BCRs) is a novel and promising alternative to the systemic suppression of humoral immunity.

Granzyme B and the Downstream Granzymes C and/or F Are Important for Cytotoxic Lymphocyte Functions [PDF]

open access: yesThe Journal of Immunology, 2005
Abstract Although the functions of granzyme A (GzmA) and GzmB are well-defined, a number of orphan granzymes of unknown function are also expressed in cytotoxic lymphocytes. Previously, we showed that a targeted loss-of-function mutation for GzmB was associated with reduced expression of several downstream orphan granzyme genes in the
