GSDME-mediated pyroptosis modulates the immunosuppressive microenvironment in lung adenocarcinoma

Acta Pharmacologica Sinica
Despite the increasing recognition of pyroptosis, particularly that involving GSDME, its precise impact on tumor prognosis and the immune microenvironment remains elusive, necessitating a comprehensive investigation in the context of lung adenocarcinoma (LUAD).

Cisplatin promotes pyroptosis of gastric cancer cells by activating GSDME

2023
ABSTRACT According to studies, numerous chemotherapeutic drugs can facilitate programmed cell death via pyroptosis. Clarifying the mechanism by which cisplatin kills gastric cancer cells is crucial for enhancing gastric cancer’s sensitivity to chemotherapy and elucidating the mechanism of drug resistance in gastric cancer.

Full-length GSDME mediates pyroptosis independent from cleavage

Nature Cell Biology
Gasdermin (GSDM) family proteins, known as the executors of pyroptosis, undergo protease-mediated cleavage before inducing pyroptosis. We here discovered a form of pyroptosis mediated by full-length (FL) GSDME without proteolytic cleavage. Intense ultraviolet-C irradiation-triggered DNA damage activates nuclear PARP1, leading to extensive formation of ...

[Research progress on GSDME-mediated cell pyroptosis resulting in GSDME type hearing loss].

Pyroptosis is a novel mode for programmed cell death discovered and confirmed in recent years. The Gasdermins (GSDMs) family is a key effector molecule mediating pyroptosis. As an important cause of extensive inflammatory damage and side effects of conventional chemotherapy drugs, anomalous pyroptosis has also been associated with hearing loss, tumor ...

RETRACTED: Streptococcus anginosus promotes gastric cancer progression via GSDME-mediated pyroptosis pathway: Molecular mechanisms of action of GSDME, cleaved caspase-3, and NLRP3 proteins

International Journal of Biological Macromolecules
Streptococcus vasculosus is a common oral and intestinal symbiotic bacteria, but it can transform into a pathogen under certain conditions, affecting the host's immune response. Studies have shown that Streptococcus vasculosus may promote tumor growth and metastasis by activating host inflammatory responses.

GSDME unlocks astrocyte-driven neurotoxicity in Alzheimer’s Disease

SUMMARY Astrocytic calcium dysregulation and reactivity precede Aβ deposition in amyloid-β deposition in Alzheimer’s disease (AD) but the neurotoxic mechanisms remain unclear. We show that GSDME acts as a switch, linking MAM-mediated calcium release to astrocyte-driven neurotoxicity.

CASP3-GSDME-mediated trophoblast pyroptosis contributes to systemic inflammation

Abstract Early-onset preeclampsia (EOPE) is associated with excessive apoptosis and inflammation, but the mechanistic link between these processes remains enigma. Here, we report elevated circulating pro-apoptotic proteins in EOPE patients at early pregnancy, along with concurrent CASP3 activation and GSDME cleavage ...
