Photoaffinity Capture Compounds to Profile the Magic Spot Nucleotide Interactomes**

open access: yesAngewandte Chemie, Volume 134, Issue 22, May 23, 2022., 2022
Molecular fishing: A family of trifunctional photoaffinity capture compounds enables the identification of putative Magic Spot Nucleotide receptors by a chemoproteomics approach. The resulting datasets cover several bacterial species, correlate with distinct nucleotide phosphorylation patterns and represent high‐value starting points for future ...

Recent functional insights into the magic role of (p)ppGpp in growth control

open access: yesComputational and Structural Biotechnology Journal, 2023
Rapid growth and survival are two key traits that enable bacterial cells to thrive in their natural habitat. The guanosine tetraphosphate and pentaphosphate [(p)ppGpp], also known as “magic spot”, is a key second messenger inside bacterial cells as well ...

The stringent response and physiological roles of (pp)pGpp in bacteria [PDF]

open access: yes, 2021
The stringent response is a stress signalling system mediated by the alarmones guanosine tetraphosphate (ppGpp) and guanosine pentaphosphate (pppGpp) in response to nutrient deprivation.

Functional Characterization of Small Alarmone Synthetase and Small Alarmone Hydrolase Proteins from Treponema denticola

open access: yesMicrobiology Spectrum, 2023
The stringent response enables bacteria to survive nutrient starvation, antibiotic challenge, and other threats to cellular survival. Two alarmone (magic spot) second messengers, guanosine pentaphosphate (pppGpp) and guanosine tetraphosphate (ppGpp ...

A rust‐fungus Nudix hydrolase effector decaps mRNA in vitro and interferes with plant immune pathways

open access: yesNew Phytologist, Volume 239, Issue 1, Page 222-239, July 2023., 2023
Summary To infect plants, pathogenic fungi secrete small proteins called effectors. Here, we describe the catalytic activity and potential virulence function of the Nudix hydrolase effector AvrM14 from the flax rust fungus (Melampsora lini). We completed extensive in vitro assays to characterise the enzymatic activity of the AvrM14 effector ...

Forming and waking dormant cells: The ppGpp ribosome dimerization persister model

open access: yesBiofilm, 2020
Procaryotes starve and face myriad stresses. The bulk population actively resists the stress, but a small population weathers the stress by entering a resting stage known as persistence.

The Borrelia burgdorferi RelA/SpoT Homolog and Stringent Response Regulate Survival in the Tick Vector and Global Gene Expression during Starvation. [PDF]

open access: yesPLoS Pathogens, 2015
As the Lyme disease bacterium Borrelia burgdorferi traverses its enzootic cycle, alternating between a tick vector and a vertebrate host, the spirochete must adapt and persist in the tick midgut under prolonged nutrient stress between blood meals.

Homology Model of a Catalytically Competent Bifunctional Rel Protein

open access: yesFrontiers in Molecular Biosciences, 2021
Bacteria have developed different bet hedging strategies to survive hostile environments and stressful conditions with persistency being maybe the most elegant yet still poorly understood one. Persisters’ temporary tolerance to antibiotic treatment hints

Robotic-Assisted Capture-Systematic Evolution of Ligands by Exponential Enrichment of RNA Aptamers Binding to Small Molecules. [PDF]

open access: yesChembiochem
This study demonstrates the rapid selection of RNA aptamers that can target various small molecules through a newly established automated capture‐systematic evolution of ligands by exponential enrichment technique. Throughout the study, this method is applied to aptamer selection for several small molecules, resulting in aptamers that bind with high ...

c‐di‐GMP regulates activity of the PlzA RNA chaperone from the Lyme disease spirochete

open access: yesMolecular Microbiology, Volume 119, Issue 6, Page 711-727, June 2023., 2023
PlzA is a c‐di‐GMP‐binding protein crucial for adaptation and survival of Borrelia burgdorferi, the etiologic agent of Lyme disease, as it cycles between its vertebrate host and tick vector. We report that PlzA has RNA chaperone activity that is regulated by c‐di‐GMP.
