Membrane recognition and dynamics of the RNA Degradosome [PDF]

open access: yes, 2015
RNase E, which is the central component of the multienzyme RNA degradosome, serves as a scaffold for interaction with other enzymes involved in mRNA degradation including the DEAD-box RNA helicase RhlB.

Rab14 regulates the transport of human papillomavirus to the trans‐Golgi network for infectious cell entry

open access: yesFEBS Letters, EarlyView.
This study reveals that the small GTPase Rab14 is necessary for human papillomavirus (HPV) infection and plays an essential role in the transport of virions to the trans‐Golgi network (TGN). HPV in the early endosome (EE), which harbors GTP‐bound Rab14, is transported to the TGN through the switch of Rab14 from its GTP‐bound to GDP‐bound form.

The RNA Polymerase II-associated factor 1 complex represses small-RNA-mediated heterochromatin formation and gene silencing [PDF]

open access: yes, 2015
RNAi interference (RNAi) is a highly conserved regulatory mechanism employed by almost all Eukaryotes. With RNAi organisms can modulate the expression of endogenous genes and protect the integrity and identity of their genomes.

Antisense pairing and SNORD13 structure guide RNA cytidine acetylation

open access: yes, 2022
International audienceN4-acetylcytidine (ac4C) is an RNA nucleobase found in all domains of life. Establishment of ac4C in helix 45 (h45) of human 18S ribosomal RNA (rRNA) requires the combined activity of the acetyltransferase NAT10 and the box C/D ...

A viral guide RNA delivery system for CRISPR-based transcriptional activation and heritable targeted DNA demethylation in Arabidopsis thaliana.

open access: yesPLoS Genetics, 2020
Plant RNA viruses are used as delivery vectors for their high level of accumulation and efficient spread during virus multiplication and movement. Utilizing this concept, several viral-based guide RNA delivery platforms for CRISPR-Cas9 genome editing ...

Shapeshifting RNAs guide innate immunity [PDF]

open access: yesJournal of Biological Chemistry, 2018
The innate immune system can distinguish between RNAs of viral and cellular origin, but the basis for this discrimination is not known. A new paper by Calderon and Conn demonstrates that conformational plasticity determines the ability of one RNA sequence to bind to and activate the pattern recognition receptor OAS1/RNase L. In identifying a novel mode

The ubiquitin‐proteasome system and autophagy as guardians of the cellular proteome

open access: yesFEBS Letters, EarlyView.
This Perspective covers the three principles governing the crosstalk between the ubiquitin‐proteasome system and autophagy in cellular proteostasis: (1) a shared ubiquitin code routing substrates via shuttle factors or autophagy receptors; (2) spatial compartmentalization into phase‐separated degradation hubs and organelle‐specific modules (exemplified

A large-scale chemical modification screen identifies design rules to generate siRNAs with high activity, high stability and low toxicity [PDF]

open access: yes, 2009
The use of chemically synthesized short interfering RNAs (siRNAs) is currently the method of choice to manipulate gene expression in mammalian cell culture, yet improvements of siRNA design is expectably required for successful application in vivo ...

Ubiquitination of secretory granules promotes their crinophagic degradation in Drosophila

open access: yesFEBS Letters, EarlyView.
Ubiquitination of secretory granules in Drosophila larval salivary glands is a critical molecular trigger for crinophagy, the lysosomal degradation of unreleased, or low‐quality granules. The E3 ubiquitin ligase Cnot4 is recruited to the surface of secretory granules to induce crinophagy.
