Human kidney-derived hematopoietic stem cells can support long-term multilineage hematopoiesis
International audienceLong-term multilineage hematopoietic donor chimerism occurs sporadically in patients who receive a transplanted solid organ enriched in lymphoid tissues such as the intestine or liver. There is currently no evidence for the presence

Derivation and characterization of retinal pigment epithelium from urine‐derived iPSCs
Age‐related macular degeneration causes vision loss via RPE dysfunction and loss. Traditional iPSC therapies rely on invasive biopsies, limiting scalability. Here, we utilize urine‐derived stem cells as an accessible source to generate u‐iPSCs, successfully differentiated into pigmented RPE. This “Urine‐to‐Retina” platform provides a promising path for

Therapeutic adenine base editing of human hematopoietic stem cells
Here, Liao and colleagues apply adenine base editor ABE8e and its PAM-less variant ABE8e-SpRY to β-thalassemia patient hematopoietic stem cells in the form of ribonucleoprotein complexes, resulting in efficient long-term editing and β-thalassemia ...

Aging Is a Key Driver for Adult Acute Myeloid Leukemia
Acute myeloid leukemia (AML) is a classical age‐related hematologic malignancy, and a key driver of AML is aging, which profoundly regulates intrinsic factors such as genomic instability, epigenetic reprogramming, and metabolic dysregulation, and alters bone marrow microenvironment.

DNMT3b's Role in Hematopoietic Stem Cells [PDF]
Hematopoiesis proceeds from a bone marrow resident population of stem cells responsible for generation of all lineages within the blood. Distinct molecular programs within hematopoietic stem cells regulate maintenance of this population under homeostatic

Mutant NPM1 in Acute Myeloid Leukemia Initiation and Maintenance
NPM1 mutations drive acute myeloid leukemia by acting as neomorphic transcriptional regulators that cooperate with Menin–MLL and XPO1 to sustain HOX/MEIS1 expression and block differentiation. Targeting these mutant‐specific transcriptional dependencies provides a rational therapeutic strategy for NPM1‐mutated AML.

Effects of Participation in Volunteer Activities on CD34+ Hematopoietic Stem Cells
Aging populations face health challenges, and volunteering may improve health outcomes. This study investigated the relationship between volunteering, CD34+ hematopoietic stem cells, and health indicators in older adults.

Role of Flk-1 in mouse hematopoietic stem cells
It was reported that human hematopoietic stem cells in bone marrow were restricted to the CD34+KDR+ cell fraction. We found that expression levels of Flk-1, a mouse homologue of KDR, were low or undetectable in mouse Lin−c-Kit+Sca-1+CD34low/− cells as ...
