Tau‐targeting active immunotherapy slows progression and reduces pathology in mouse models of tauopathy

The efficacy of the novel anti‐tau active immunotherapy, p5555kb, was tested using two mouse models of tau pathology. p5555kb inoculation increased the survival rate and reduced tau pathology in tau‐overexpressing P301L mice and decreased tau seeding in the brains of C57BL/6 mice injected with human‐purified Alzheimer's disease tau.

ヘパラン硫酸のコアタンパク質に脂質が共有結合すると、これらの遺伝子産物と細胞膜間に新しい分子結合が出来ることになる。プロテオグリカンは糖脂質化することで細胞頂上部へのふるい分けと標的となる膜への移動を行ない、細胞表面のプロテオグリカンが酵素反応により敏速に遊離される機構が出来上がる。プロテオグリカンが脂肪酸でアシル化されることは、疎水的な相互作用と、恐らく受容体を介した結合を仲介し、細胞内のプロテオグリカン/プロテオグリカン同士の相互作用の新しい機構を提供し、代謝回転に直接的な役割を果たすかも知れない。脂質を介した繋留に必要となる微細構造上の要求性や複合的なな酵素反応装置が同定出来ると、プロテオグリカンのこの新しい翻訳後の修飾が機能的に重要であることが明らかとなるであろう。

The extracellular matrix glycoprotein opticin is a small leucine-rich repeat proteoglycan/protein family member that was discovered associated with vitreous humor collagen fibrils.
