A generally conserved response to hypoxia in iPSC-derived cardiomyocytes from humans and chimpanzees

open access: yeseLife, 2019
Despite anatomical similarities, there are differences in susceptibility to cardiovascular disease (CVD) between primates; humans are prone to myocardial ischemia, while chimpanzees are prone to myocardial fibrosis.

Whole‐Genome Sequencing Pilot of the Central Asian Genomic Diversity Project Reveals Distinct Histories, Adaptation, and Introgression

open access: yesAdvanced Science, EarlyView.
As a pilot phase of the Central Asian Genomic Diversity Project, whole‐genome sequencing of 166 individuals from 20 Central Asian and Afghan Hazara populations reveals fine‐scale substructure shaped by repeated trans‐Eurasian migration and admixture. Integrated analyses uncover post‐admixture adaptation, archaic introgression, and medically relevant ...

Mechanisms and Therapeutic Potential of Human Cardiomyocyte Proliferation

open access: yesJournal of Cardiovascular Development and Disease
The limited capacity for cardiomyocyte proliferation in the adult human heart restricts its ability to recover from injury. Building on discoveries in regenerative model systems, such as zebrafish and neonatal mice, reactivation of a latent potential for cardiomyocyte proliferation is a strategy to promote therapeutic heart regeneration.

Intermittent Fasting Restores Cardiac Lipid Homeostasis in Diabetic Cardiomyopathy in Association With Akkermansia Muciniphila and 1‐methyl‐L‐histidine

open access: yesAdvanced Science, EarlyView.
Intermittent fasting reshapes the gut microbiota in diabetic cardiomyopathy, restoring Akkermansia muciniphila and the microbiota‐associated metabolite 1‐methyl‐L‐histidine. This shift is linked to improved cardiac lipid homeostasis, reduced lipid peroxidation, and attenuated myocardial injury, highlighting a gut microbiota–metabolite–lipid axis in ...

QRICH1 Disrupts Endoplasmic Reticulum Homeostasis and Amplifies NF‐κB Signaling in Periodontal Ligament Stem Cells to Exacerbate Diabetic Periodontitis

open access: yesAdvanced Science, EarlyView.
QRICH1 has been established as a key factor contributing to impaired osteogenic potential and accelerated apoptosis of PDLSCs in diabetic periodontitis. QRICH1 not only significantly enhances UPR‐associated apoptotic signaling but also amplifies NF‐κB‐mediated inflammatory responses. Its inhibition restores osteogenic capacity and reduces alveolar bone

Characterization of cytoskeleton features and maturation status of cultured human iPSC-derived cardiomyocytes

open access: yesEuropean Journal of Histochemistry, 2017
Recent innovations in stem cell technologies and the availability of human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) have opened new possibilities for studies and drug testing on human cardiomyocytes in vitro.

A Novel Pak1 Activator Ameliorates ER Stress for HFpEF Therapy

open access: yesAdvanced Science, EarlyView.
Chronic metabolic stress is a major contributor to HFpEF progression. Under prolonged metabolic stress, Pak1 activity becomes impaired, contributing to disrupted ER proteostasis, cardiomyocyte apoptosis, fibrosis, and diastolic dysfunction. Mechanistically, Pak1 overexpression activates the ERK1/2–MNK1–eIF4E signaling axis, promotes translational ...

Programmable Paracrine‐Mimetic Microneedle System for Temporal Regulation of Cardiac Repair

open access: yesAdvanced Science, EarlyView.
Temporal signaling is encoded into a programmable paracrine‐mimetic microneedle patch via tris(2,2'‐bipyridyl)dichlororuthenium(II) hexahydrate/sodium persulfate‐tuned hydrogel microparticles that deliver TGF‐β, IGF‐1, and VEGF over phase‐matched 7‐, 14‐, and 28‐day windows.

Nucleoside‐Modified mRNA Encoding Alpha‐Galactosidase A Ameliorates Fabry Disease Phenotypes in Human IPSC‐Derived Cardiomyocytes

open access: yesAdvanced Science, EarlyView.
Human iPSC‐derived Fabry cardiomyocytes exhibited broad transcriptional dysregulation, apoptosis, mitochondrial dysfunction, impaired reactive oxygen species handling, altered contractility, and abnormal calcium transient decay, potentially mediated by phospholamban hyperphosphorylation.
