Hydroxyl radical footprinting

2000
Abstract Hydroxyl radicals have now been used for more than 15years to probe the structure of various macromolecules. They have also been widely used to footprint DNA-protein and RNA-protein complexes (1, 2). Hydroxyl radicals are w1y small, comparable in size to a water molecule, are most commonly generated from FeEDTA in solution by

Hydroxyl radical-scavenging property of indomethacin

Molecular and Cellular Biochemistry, 1994
The ability of indomethacin to scavenge hydroxyl radical (.OH) using high pressure liquid chromatography (HPLC) was investigated. .OH radical was generated by photolysis of H2O2 (1.5-10 mmoles/L) with UV light and was trapped with salicyclic acid (500 nmoles). H2O2 produced .OH in a concentration-dependent manner as estimated by .OH adduct products 2,3-

Scavenging of hydroxyl radical by catecholamines

Luminescence, 2012
ABSTRACTThe direct effects of the four catecholamines (CATs), adrenaline (A), noradrenaline (NA), dopamine (D) and isoproterenol (I), on free radicals were investigated using the free radical 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH•) and hydroxyl radial (HO•).

Hydroxyl radical induced degradation of ibuprofen

Science of The Total Environment, 2013
Pulse radiolysis experiments were used to characterize the intermediates formed from ibuprofen during electron beam irradiation in a solution of 0.1mmoldm(-3). For end product characterization (60)Co γ-irradiation was used and the samples were evaluated either by taking their UV-vis spectra or by HPLC with UV or MS detection.

Hydroxyl Radical Generation by Photosystem II

Biochemistry, 2004
The photogeneration of hydroxyl radicals (OH(*)) in photosystem II (PSII) membranes was studied using EPR spin-trapping spectroscopy. Two kinetically distinguishable phases in the formation of the spin trap-hydroxyl (POBN-OH) adduct EPR signal were observed: the first phase (t(1/2) = 7.5 min) and the second phase (t(1/2) = 30 min). The generation of OH(

Photogeneration of hydroxyl radicals for footprinting

Analytical Biochemistry, 1992
Hydroxyl radicals yield footprints of DNA-ligand interactions that are uniform in intensity and display single base pair resolution. It is shown here that brief illumination of dilute aqueous solutions of hydrogen peroxide with a standard uv transilluminator can be used to generate hydroxyl radicals for footprinting studies.

Isotopic Spectra of the Hydroxyl Radical

The Journal of Physical Chemistry A, 2013
Rotational spectra of OH and its isotopologues have been precisely measured using high efficiency terahertz (THz) sources. The measurements are compared with existing data and are useful for global modeling. For the first time, microwave measurements of the Λ-doubling transitions of the (17)OH isotopologue are combined with THz data successfully ...

DNA Footprinting with the Hydroxyl Radical

Free Radical Research Communications, 1991
The Fenton reaction of iron(II) EDTA with hydrogen peroxide, performed in the presence of ascorbate ion, has proven to be useful as a probe of structure in DNA systems. Two aspects of this chemistry are discussed: the identity of the active DNA cleaving agent produced by this reagent, and the application of the Fenton reaction to the determination of ...
