USP14 promotes tryptophan metabolism and immune suppression by stabilizing IDO1 in colorectal cancer

open access: yes, 2022
Indoleamine 2,3 dioxygenase 1 (IDO1) is an attractive target for cancer immunotherapy. However, IDO1 inhibitors have shown disappointing therapeutic efficacy in clinical trials, mainly because of the activation of the aryl hydrocarbon receptor (AhR ...

Activation of indoleamine 2,3-dioxygenase in patients with scrub typhus and its role in growth restriction of Orientia tsutsugamushi. [PDF]

open access: yesPLoS Neglected Tropical Diseases, 2012
BACKGROUND: Our earlier genome-wide expression study revealed up-regulation of a tryptophan-catabolizing enzyme, indoleamine 2,3-dioxygenase (IDO1), in patients with scrub typhus.

Monocyte inflammation and adaptive immune dysfunction in severe Klebsiella pneumoniae pneumonia

open access: yesiMeta, EarlyView.
Severe Klebsiella pneumoniae pneumonia combines intense inflammation with failure to control infection, yet the cellular basis is unclear. Using a severity‐focused single‐cell atlas of 708,894 peripheral blood mononuclear cells from 100 individuals, complemented by plasma protein measurements and flow cytometry, we show that disease severity redirects ...

Discovery and evaluation of inhibitors to the immunosuppressive enzyme indoleamine 2,3-dioxygenase 1 (IDO1):Probing the active site-inhibitor interactions

open access: yes, 2017
High expression of the immunosuppressive enzyme, indoleamine 2,3-dioxygenase 1 (IDO1) for a broad range of malignancies is associated with poor patient prognosis, and the enzyme is a validated target for cancer intervention.

Organoid Coculture Models for Cancer Research and Immunotherapy

open access: yesMed Research, EarlyView.
This review summarizes organoid coculture platforms, including submerged Matrigel culture, air–liquid interface, microfluidic/organoid‐on‐a‐chip, and 3D bioprinting, for reconstructing the tumor microenvironment to study tumor–stroma/immune crosstalk, evaluate immunotherapies, and enable drug screening.

Current Advances of Treatments, Therapeutic Targets, Biomarkers, Novel Drugs, and Machine Learning for Hypopharyngeal Squamous Cell Carcinoma

open access: yesWorld Journal of Otorhinolaryngology - Head and Neck Surgery, EarlyView.
ABSTRACT Hypopharyngeal squamous cell carcinoma (HSCC) is an aggressive subtype of head and neck squamous cell carcinoma with insidious onset, early metastasis, and dismal prognosis. Conventional multimodal therapy achieves limited survival benefit, highlighting an urgent need for refined precision strategies.

Kaempferol Protects Intestinal Health in Chinese Forest Musk Deer Possibly by Regulating Intestinal Microbiota and Inhibiting the NF‐κB/NLRP3 Signaling Pathway

open access: yesIntegrative Zoology, EarlyView.
Kaempferol modulates the tryptophan metabolism pathway by increasing the abundances of Christensenellaceae R7 group, Bacteroides, and Blautia and reducing that of Rikenellaceae RC9 gut group, thereby significantly increasing the levels of ILA and IAA. This process inhibits the activation of NF‐κB/NLRP3 signaling pathway, reduces pro‐inflammatory factor

Tryptophan deficiency induced by indoleamine 2,3‐dioxygenase 1 results in glucose transporter 1‐dependent promotion of aerobic glycolysis in pancreatic cancer

open access: yesMedComm
Indoleamine 2,3‐dioxygenase 1 (IDO1), the key enzyme in the catabolism of the essential amino acid tryptophan (Trp) through kynurenine pathway, induces immune tolerance and is considered as a critical immune checkpoint, but its impacts as a metabolism ...

Lupus Panniculitis Immunoprofiling Reveals a Cytotoxic, Type 1‐Skewed T‐Cell Response With Signs of Metabolic Alteration

open access: yesAllergy, EarlyView.
Imaging mass cytometry shows lupus panniculitis lesions are dominated by cytotoxic and skin‐homing T cells with abundant macrophages and B‐cell aggregates. Spatial neighborhood analysis reveals organized leukocyte architecture, including B‐cell proximity to naïve T cells and M1 macrophages to cytotoxic T cells.

Indoleamine 2,3-dioxygenase 1 (IDO1) activity correlates with immune system abnormalities in multiple myeloma

open access: yesJournal of Translational Medicine, 2012
Background Multiple myeloma (MM) is a plasma cell malignancy with a multifaceted immune dysfunction. Indoleamine 2,3-dioxygenase 1 (IDO1) degrades tryptophan into kynurenine (KYN), which inhibits effector T cells and promote regulatory T-cell (Treg ...
