Nascent Extracellular Matrix Converts Biomaterial Cues into Cell Fate Decisions

open access: yesAdvanced Science, EarlyView.
This study uncovers a tri‐directional interplay among cells, hydrogels, and their nascent extracellular matrix. Hydrogel norbornene modification alters CD44‐mediated cell–hydrogel interactions, which shape nascent matrix deposition, composition, and spatial organization.

Tissue inhibitors of matrix metalloproteinases in cancer.

open access: yes, 1999
Tissue inhibitors of metalloproteinases (TIMPs) play a key regulatory role in the homeostasis of the extracellular matrix (ECM) by controlling the activity of matrix metalloproteinases (MMPs). Some TIMPs have a second function as well, unrelated to their

Bioenergetic Materials for Tissue Regeneration: Modulating Metabolism to Promote Cellular Anabolism

open access: yesAdvanced Science, EarlyView.
Bioenergetic materials (BEMs) modulate cellular metabolism to promote tissue regeneration. Their principal mechanisms of action include metabolic substrate supplementation, metabolic enzyme modulation, organelle transfer, electrical current regulation, redox homeostasis modulation, and oxygen tension modulation.

Transfer RNA‐Derived Small RNA‐5’tiRNAGln Drives HNRNPC Phase Separation to Promote Breast Cancer Bone Metastasis by Regulating IGF1R

open access: yesAdvanced Science, EarlyView.
This study reveals that 5'tiRNA‐Gln interacts with hnRNPC to promote IGF1R liquid‐liquid phase separation, which drives ERK1/2 signaling activation, EMT, and breast cancer bone metastasis. These findings highlight a novel mechanism of breast cancer bone metastasis and potential therapeutic target in metastatic breast cancer.

Versatile Microgel Platform for Intervertebral Disc Degeneration Therapy: Targeting Pericyte‐Mediated Fibrosis and Protecting Nucleus Pulposus Cells

open access: yesAdvanced Science, EarlyView.
The engineered MMS@TRP microgel enables targeted, acid‐responsive delivery of a miR‐21 inhibitor to pericytes. It effectively suppresses disc fibrosis by inhibiting TGF‐β signaling while protecting nucleus pulposus cells via sustained, antioxidant tannic acid release.

Neonatal Sevoflurane Exposure Induces Long‐Term Cognitive Impairment via Epigenetically Mediated MMP9 Activation and Perineuronal Net Disruption in the Hippocampal CA2

open access: yesAdvanced Science, EarlyView.
Hippocampal CA2 perineuronal nets ensheathing excitatory pyramidal neurons are selectively vulnerable to repeated neonatal sevoflurane exposure. Epigenetic upregulation of MMP9 via reduced H3K27me3 disrupts PNN integrity, impairs BDNF–TrkB signaling, and causes synaptic protein loss and dendritic spine deficits, leading to persistent social and spatial

A Spatiotemporal Single‐Cell Atlas Uncovers Dysregulated ECM Dynamics and Septal Remodeling Arrest in Human Ventricular Septal Defects

open access: yesAdvanced Science, EarlyView.
Integrating single‐cell and spatial transcriptomics reveals human ventricular septal defect (VSD) as a disease of microenvironmental insufficiency and structural remodeling arrest. Attenuated endothelial‐fibroblast crosstalk, alongside downregulated THBS1/TGF‐β1 and MMP2 signaling networks, impairs the endothelial‐to‐mesenchymal transition (EndoMT) and

ACAT1‐Mediated SP1‐K694 Lactylation Promotes M1 Macrophage Polarization via TREM1 Transcription in COPD

open access: yesAdvanced Science, EarlyView.
Upon cigarette smoke exposure(CSE), LDHA expression is increased in lung macrophages, promoting the conversion of pyruvate to lactate and elevating the lactylation level of SP1 at the K694 site. This, in turn, enhances SP1 enrichment at the TREM1 promoter, increases TREM1 expression, promoted M1 macrophage polarization, and triggers the secretion of ...

Long‐Term Human Skin Platform for Modeling Chronic Inflammation, Environmental Stress, and Therapeutic Intervention

open access: yesAdvanced Science, EarlyView.
An advanced, long‐term human skin platform maintains native skin structure and key tissue functions for up to four weeks. The platform models chronic inflammation, aging‐related skin changes, and UVB damage, enabling drug and topical interventions—including anti‐inflammatory therapy, senolytic treatment, and sunscreen—to be evaluated in intact human ...
