Persistent Innate Immune Stimulation Results in IRF3-Mediated but Caspase-Independent Cytostasis
Persistent virus infection continuously produces non-self nucleic acids that activate cell-intrinsic immune responses. However, the antiviral defense evolved as a transient, acute phase response and the effects of persistently ongoing stimulation onto ...

Macrophage RSAD2 dually regulates CMPK2—the rate‐limiting enzyme for mitochondrial DNA synthesis—by suppressing its ubiquitination and promoting its phosphorylation via Csnk2a2 recruitment. This amplifies mtDNA production, which simultaneously activates a cGAS‐STING‐IRF3‐RSAD2 feedforward loop and the NLRP3 inflammasome, driving a self‐sustaining ...

Suppressing FXR promotes antiviral effects of bile acids via enhancing the interferon transcription
Bile acids (BAs) are natural metabolites in mammals and have the potential to function as drugs against viral infection. However, the limited understanding of chenodeoxycholic acid (CDCA) receptors and downstream signaling, along with its lower ...

The EF-Hand Protein CALML6 Suppresses Antiviral Innate Immunity by Impairing IRF3 Dimerization
Summary: The transcription factor IRF3 is phosphorylated in response to viral infection, and it subsequently forms a homodimer and translocates into the nucleus to induce the transcription of genes important for antiviral immunity, such as type I ...

D4F‐functionalized ceria nanozyme–CasRx nanoparticles preferentially target renal macrophages through SR‐B1‐mediated uptake in ischemia–reperfusion injury. Ceria scavenges excessive ROS, while CasRx silences STING signaling, shifting macrophages toward a less inflammatory, pro‐resolving state and increasing the proportion of regulatory T cells.

Ox‐mtDNA fragments escaping from mitochondria drive robust inflammatory responses. A targeted nanoplatform simultaneously inhibits mitochondrial FEN1‐mediated mtDNA cleavage and scavenges ROS, preventing the generation of immunogenic Ox‐mtDNA fragments. The released SeNPs further promote autophagic clearance of cytosolic mtDNA. Consequently, cGAS‐STING,

Interferon Regülatör Faktör 3 (Irf3) Geninin Genetik Analizi
Interferon Regulating Factor 3 (IRF3) protein is a member of IRF (Interferon Regulating Factors) family. Most of IRFs play an important role in the constitution of responses to pathogens, differentiation hematopoietic cells, regulation of gene expression,

Of Mice and Men and Cattle: Functions of the Pneumovirus Nonstructural Proteins NS1 and NS2 in Interferon Escape [PDF]
Bovine respiratory syncytial virus (BRSV), family Paramyxoviridae, subfamily Pneumovirinae, is a major etiological agent of respiratory tract disease in calves. A peculiarity of pneumoviruses is the presence of two unique nonstructural protein genes, NS1

Interferon Regulatory Factor 3 Exacerbates the Severity of COVID-19 in Mice
CONTEXT:. Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) emerged in 2019, causing the COVID-19 pandemic. While most infected people experienced mild illness, others progressed to severe disease, characterized by hyperinflammation and ...
