AbstractA murine macrophage cell line P388D1 in in vitro culture without any specific stimulation produced both interleukin 1 (IL1) and IL1 inhibitor which inhibits mitogenic response of murine thymocytes to IL1 in the culture fluids. The factor(s) responsible for inhibiting IL1‐induced thymocyte proliferation consisted of at least two molecules ...

Rat Kupffer cells stimulated with bacterial lipopolysaccharide (LPS) produced high levels of interleukin 1 (IL‐1), as determined by thymocyte proliferation assay. Indomethacin revealed a dose‐dependent augmentation in IL‐l production, in parallel with a dose‐dependent reduction in prostaglandin E2 production by Kupffer cells.

Interferons as inhibitors of interleukin 1 induced interleukin 1 synthesis.
Lymphokine research, 1989IL-1 induces its own gene expression in cultured smooth muscle and endothelial cells and in human PBMC. IL-1-induced IL-1 may be part of a self-amplification or autocrine event in inflammation. In the present study IFN gamma consistently increased LPS-induced IL-1, but reduced the total amount of IL-1-induced IL-1 from PBMC. On a molar basis, IFN gamma

2'-Substituted chalcone derivatives as inhibitors of interleukin-1 biosynthesis
Journal of Medicinal Chemistry, 1993A series of 2'-substituted chalcone derivatives has been found to show potent inhibition of the production of IL-1 beta from human peripheral blood monocytes stimulated with lipopolysaccharide (LPS), with IC50 values in the 0.2-5.0-microM range. Some members of the series have also shown inhibition of septic shock induced in mice by injection of LPS ...

Interleukin-1 (IL-1) Inhibitors: Anakinra, Rilonacept, and Canakinumab
2016Inhibitors of interleukin-1 (IL-1) are proteins which decrease the actions of the inflammatory cytokine, IL-1. There are two general mechanisms of IL-1 inhibitors: binding to the IL-1 receptor (anakinra) or binding directly to IL-1 (rilonacept and canakinumab). They are members of a general class termed biological disease-modifying antirheumatic drugs (

Role of Interleukin‐1 Inhibitors in the Management of Gout
Pharmacotherapy: The Journal of Human Pharmacology and Drug Therapy, 2013Interleukin‐1 (IL‐1) inhibitors potentially have a role as antiinflammatory agents in refractory gout or for patients who are unable to tolerate conventional therapy, such as nonsteroidal antiinflammatory drugs (NSAIDs), colchicine, or glucocorticoids, for acute attacks.

Molecular size chromatography of urine from normal individuals showed two peaks of apparent IL-1 suppressive activities when tested by the murine thymocyte comitogenic assay (mol wt greater than 600 kD and 20 to 60 kD). However, the high molecular weight inhibitory activity disappeared if the concentration of PHA was increased during assay, and the low

Tripterygium wilfordii var. regelii which are interleukin-1 inhibitors
Phytochemistry, 1994Abstract Seven new quinoide-type diterpenes were isolated from the stems of Tripterygium wilfordii var. regelii and their structures were established on the basis of spectroscopic and chemical evidence. The isolated compounds showed extremely potent inhibitory activities against interleukin-lα and β releases for human peripheral mononticlear ...

AbstractInterleukin‐1 (IL‐1) has been shown to regulate glycosaminoglycan (GAG) synthesis. We therefore investigated whether an IL‐1 inhibitor or IL‐6 modulates IL‐1 biologic activities in human synovial cells and cultured articular cartilage. We found that in the presence of a constant amount of IL‐1β, stimulation of hyaluronic acid (HA) synthesis by ...

Interleukin-1 induces chondrocyte protease production: The development of collagenase inhibitors
Agents and Actions, 1987Supernatants from the P388D1 macrophage cell line as well as human interleukin-1 (IL-1) stimulated primary rabbit articular chondrocytes to produce collagen- (C-ase) and proteoglycan- (PG-ase) degrading proteases. The P388D1 derived factor had a molecular weight of 16,000-20,000 and a pI of 4.5-5.0.
