Upon VZV entry, the viral kinase ORF47p, which is part of the VZV tegument, is released in the cytoplasm where it can encounter IRF3 and induce its atypical phosphorylation leading to an up-shift in the electrophoretic mobility of IRF3.

Tian et al. reveal that FH K112 lactylation is elevated after TBI, leading to fumarate accumulation, mitochondrial damage, mtDNA release and amplified neuroinflammation. AARS2 and SIRT3 act as the “writer” and “eraser” for FH K112 lactylation. The peptide Pep‐K112 targeting this site alleviates brain injury, highlighting a potential therapeutic target.

TP53 Loss Elevates NF‐κB‐IFN‐β‐MHC‐Ia Signaling to Promote NK Cell Resistance in Osteosarcoma
TP53 loss in a transforming or osteosarcoma cell promotes cytosolic DNA accumulation, activating NF‐κB‐dependent IFN‐β production. Autocrine IFN‐β signaling increases cell‐surface HLA‐Ia expression, strengthens inhibitory KIR signaling in natural killer cells, and thereby enables the affected cell to evade NK cell‐mediated cytotoxicity.

Pathological tissue rigidity mechanoprimes microglia by enhancing actin cytoskeleton–nucleus coupling and chromatin opening at rigidity‐responsive cis‐regulatory elements (mechanoCREs).
