Microglial Deubiquitinase OTUD7B Stabilizes STAT3 to Promote Neuroinflammation and Cognitive Decline in Alzheimer's Disease

open access: yesAdvanced Science, EarlyView.
. ABSTRACT Neuroinflammation driven by microglial activation is a defining feature of Alzheimer's disease (AD), yet the molecular mechanisms sustaining this proinflammatory state remain unclear. Here, we identify the deubiquitinase OTUD7B as a critical regulator of microglial activation and AD pathology.

RNF26 regulates IRF3 stability.

open access: yes, 2014
(A) Effects of RNF26 knockdown on IRF3 level. In the left panel, whole cell lysates of THP-1-RNF26-RNAi or control cells (1×106) were analyzed by immunoblots with the indicated antibodies.

Engineering Microbial Particles for Next‐Generation Biomedical Platforms

open access: yesAdvanced Science, EarlyView.
Microbe‐derived particles (MDPs), which include extracellular vesicles, outer membrane vesicles, inclusion bodies, polysaccharide particles, and virus‐like particles, represent a rapidly expanding category of bioinspired nanomaterials. With their natural origin, intrinsic biocompatibility, and highly programmable functionality, MDPs serve as a ...

IRF3 Negatively Regulates Toll-Like Receptor-Mediated NF-κB Signaling by Targeting TRIF for Degradation in Teleost Fish

open access: yesFrontiers in Immunology, 2018
NF-κB signaling is tightly regulated and essential to innate and adaptive immune responses, its regulatory mechanism remains unclear in various organisms, especially teleosts.

Harmonizing Rigidity and Flexibility: Embedding COF Quantum Dots Into Extracellular Matrix Gel as Carbon Monoxide Depot for Acoustically Triggered Time‐Programmable Anti‐Infective Therapy

open access: yesAdvanced Science, EarlyView.
A rigid‑flexible integrated CO depot is engineered by embedding rigid COFQDs into a flexible ECM gel network. The rigid framework enables high‑capacity CO storage and sonodynamic bacterial killing, while the flexible ECM gel controls sustained CO release, collectively eradicating multidrug‑resistant infection and promoting scarless healing.

Cellular localization of the herpes simplex virus ICP0 protein dictates its ability to block IRF3-mediated innate immune responses. [PDF]

open access: yesPLoS ONE, 2010
Interferon regulatory factor 3 (IRF3) is important for innate antiviral responses; accordingly, many viruses target and inactivate IRF3. The ability of the Herpes simplex virus type 1 (HSV-1) immediate early protein ICP0 to inhibit IRF3 is controversial ...

The Non-transcriptional Function of IRF3 Dynamically Regulates Immune Cell Populations in Acute on Chronic Ethanol in Mice

open access: yesRevista Cubana de Investigaciones Biomédicas, 2020
Introduction: Interferon regulatory factor 3 (IRF3) is a transcription factor mediating anti-viral responses, yet recent evidence indicates that IRF3 also has critical non-transcriptional functions, including activating RIG-I-like receptors-induced IRF-3-

TRIM26 promotes IRF3 ubiquitination and proteasomal degradation.

open access: yes, 2015
(A) Lysates from mice peritoneal macrophages stimulated with LPS or infected with SeV for indicated time periods were subjected to immunoprecipitation with anti-IRF3 antibody followed by western blot analysis with anti-TRIM26 antibody.

AI Designed Conformation Locking Peptides Target STING to Restore Diabetic Wound Healing

open access: yesAdvanced Science, EarlyView.
A generative AI pipeline identifies SCP‐1, a conformation‐locking peptide that stabilizes inactive STING. Incorporated into a dual‐responsive hydrogel for localized, MMP‐9‐triggered release, SCP‐1 suppresses inflammation, promotes reparative macrophage polarization, and accelerates diabetic wound healing.
