Macrolide and macrolide-related antibacterial agents
マクロライド系抗菌薬は,グラム陽性菌および非定型病原体に対する抗菌力が強く,呼吸器疾患の治療薬として汎用されてきた.我が国では,14員環,15員環および16員環マクロライド系およびケトライド系抗菌薬が上市している.エリスロマイシンの欠点を克服するための創薬研究から,ニューマクロライドと呼ばれるマクロライド系抗菌薬のロキシスロマイシン,クラリスロマイシンおよびアジスロマイシンが半合成された.その後,耐性肺炎球菌に対する抗菌活性が増強された,ケトライド系抗菌薬のテリスロマイシンが半合成された.本稿では主として,14,15員環マクロライド系およびケトライド系を中心とした抗菌薬の創薬・開発研究の歴史ならびに抗菌作用の特徴を概説した.
